摘  要:目的利用两样本双向孟德尔随机化(MR)法研究维生素D和慢性丙型肝炎(CHC)的潜在因果关系。方法利用全基因组关联研究(GWAS)汇总数据集,运用逆方差加权法(IVW)、MR-Egger回归法、加权中位数法、简单模式法和加权模式法进行两样本MR分析。采用IVW和MR-Egger回归法检验异质性,采用MR-Egger回归法评估基因水平多效性,采用留一法评估结果的稳定性。结果IVW结果显示,维生素D对CHC无因果影响[OR=1.122,95%CI(0.841~1.498),P=0.434];反之,CHC对维生素D也无因果影响[OR=0.988,95%CI(0.961~1.015),P=0.369]。其他MR方法与IVW结果基本一致;未发现异质性与基因水平多效性(P>0.05)。结论在一个大型欧洲队列中,未发现维生素D与CHC之间存在因果关系。Objective To study the potential causal relationship between vitamin D and chronic hepatitis C(CHC)using two-sample bidirectional Mendelian randomization(MR).Methods The data set was summarized by genome-wide association study(GWAS),and two-sample MR analysis was performed using inverse-variance weighted(IVW),MR-Egger regression method,weighted median method,simple mode method and weighted mode method.IVW and MR-Egger regression methods were used to test heterogeneity,MR-Egger regression method was used to evaluate gene-level pleiotropy,and leave-one-out method was used to evaluate the stability of the results.Results The results of IVW showed that vitamin D had no causal effect on CHC[OR=1.122,95%CI(0.841-1.498),P=0.434];on the contrary,CHC had no causal effect on vitamin D[OR=0.988,95%CI(0.961-1.015),P=0.369].Other MR methods were basically consistent with IVW results;no heterogeneity and pleiotropic at the gene level were found(P>0.05).Conclusion In a large European cohort,no causal relationship between vitamin D and CHC was found.
