摘 要:目的分析盐酸凯普拉生对幽门螺杆菌体外抗菌活性的影响。方法采用琼脂稀释法测定盐酸凯普拉生三联疗法(凯普拉生+克拉霉素+阿莫西林)对幽门螺杆菌标准菌株ATCC43504和克拉霉素耐药菌株YM-2024-D2-009的最低抑菌浓度(MIC);固定三联疗法药物浓度比,加入不同浓度的枸橼酸铋钾,形成四联疗法,判读四联疗法各浓度药物的体外抗菌结果,并比较三联疗法和四联疗法的抗菌活性。结果三联疗法对幽门螺杆菌标准菌株ATCC43504的MIC为凯普拉生∶克拉霉素∶阿莫西林=0.000975000000∶0.024375000000∶0.048750000000μg/mL;添加枸橼酸铋钾后,四联疗法的药物浓度为三联疗法MIC的1/2~1/32。三联疗法对幽门螺杆菌克拉霉素耐药菌株YM-2024-D2-009的MIC为凯普拉生∶克拉霉素∶阿莫西林=0.003900000∶0.097500000∶0.195000000μg/mL;添加枸橼酸铋钾后,四联疗法的药物浓度为三联疗法MIC的1/2。结论盐酸凯普拉生三联疗法和四联疗法均能有效抑制幽门螺杆菌标准菌株和克拉霉素耐药菌株的生长,其中四联疗法效果优于三联疗法。Objective To analyze the effect of Keverprazan Hydrochloride on the antibacterial activity of helicobacter pylori in vitro.Methods The agar dilution method was used to determine the minimum inhibitory concentration(MIC)of the triple therapy of Keverprazan Hydrochloride(Keverprazan+Clarithromycin+Amoxicillin)on helicobacter pylori standard strain ATCC43504 and Clarithromycin-resistant strain YM-2024-D2-009.The concentration ratio of triple therapy drugs was fixed,and different concentrations of Bismuth Potassium Citrate were added to form a quadruple therapy in order to evaluate and distinguish the in vitro antibacterial results of each concentration of quadruple therapy drugs.The antibacterial activity was compared between triple therapy and quadruple therapy.Results The MIC of triple therapy for helicobacter pylori standard strain ATCC43504 was as follows Keverprazan:Clarithromycin:Amoxicillin was 0.000975000000:0.024375000000:0.048750000000μg/mL;after adding Bismuth Potassium Citrate,the drug concentration of quadruple therapy was 1/2 to 1/32 of the MIC of triple therapy.The MIC of triple therapy for helicobacter pylori Clarithromycin-resistant strain YM-2024-D2-009 was as follows Keverprazan:Clarithromycin:Amoxicillin was 0.003900000:0.097500000:0.195000000μg/mL;after adding Bismuth Potassium Citrate,the drug concentration of quadruple therapy was 1/2 of the MIC of triple therapy.Conclusions Both triple therapy and quadruple therapy of Keverprazan Hydrochloride can effectively inhibit the growth of helicobacter pylori standard strains and Clarithromycinresistant strains,and the effect of quadruple therapy is better than that of triple therapy.
