摘 要:采集朽木下10 cm土样,用刚果红染色法对菌株进行初筛,使用复筛培养基进一步筛选得到2株木质纤维素降解菌ysy3和ysy4。菌株ysy3鉴定为芽孢杆菌(Bacillus sp.),菌株ysy4鉴定为链霉菌(Streptomyces sp.)。菌株ysy3以玉米芯为底物在37℃进行酶活力的测定,结果表明菌株ysy3的酶活力均呈现先增长后下降的趋势,第4天达到酶活性最大值,滤纸酶活力为0.047 U/mL;内切葡聚糖酶活力为0.127 U/mL;木聚糖酶活力为9.81 U/mL。菌株ysy4分别在28℃、37℃、45℃进行酶活力测定,结果显示在28℃培养条件下,酶活性较高,在第13天达到最大值,滤纸酶活力为0.065 U/mL;内切葡聚糖酶活力为0.149 U/mL;木聚糖酶活力为15.09 U/mL。菌株ysy3,ysy4纤维素降解能力较强,滤纸酶活性和木聚糖酶活性均较高,具有生物降解开发应用价值。Soil samples were collected from under decayed wood at a depth of 10 cm.The Congo red staining method was used for the initial screening of the strains,and further selection using a re-screening culture medium resulted in the isolation of two strains of lignocellulose-degrading bacteria,ysy3 and ysy4.Strain ysy3 was identified as Bacillus sp.,and strain ysy4 was identified as Streptomyces sp..The strain ysy3 was tested for enzyme activity using corncob as the substrate at 37℃.The results indicated that the enzyme activity of strain ysy3 showed a trend of first increasing and then decreasing,reaching its maximum enzyme activity on the 4th day.The filter paper enzyme activity was 0.047 U/mL;the endo-glucanase activity was 0.127 U/mL;and the xylanase activity was 9.81 U/mL.The strain ysy4 was subjected to enzyme activity measurement at 28℃,37℃,and 45℃.The results showed that under the cultivation conditions of 28℃,the enzyme activity was relatively high,reaching its maximum on the 13th day,with filter paper enzyme activity at 0.065 U/mL;endo-glucanase activity at 0.149 U/mL;and xylanase activity at 15.09 U/mL.Strains ysy3 and ysy4 have strong cellulose degradation capabilities,with high filter paper enzyme activity and xylanase activity,indicating their potential value for bioremediation and application development.
