摘  要:点击化学反应在多个研究领域具有独特的作用,但作为本科生教学实验则鲜有报道。本实验以叠氮乙酸乙酯为原料,经过水解、活化、交联等步骤,构建了叠氮修饰的牛血清蛋白,随后与炔丙基荧光素进行点击化学反应,实现了蛋白荧光标记,并利用凝胶电泳对标记蛋白进行了表征。本实验将多学科基础知识有机融合,通过多步实验,直观展示了其在研究中的应用,有助于本科生更好地理解这些知识,并与实践相结合,培养其综合能力。Click reactions play a unique role in various research fields.However,there are few reports of this being used as an undergraduate teaching experiment.In this experiment,ethyl azidoacetate was used as the starting material,and through steps of hydrolysis,activation,and conjugation,azide-modified bovine serum albumin was constructed.This was followed by the click reaction with propargylamine-derived fluorescein to achieve protein fluorescent labeling.The labeled protein was then separated and characterized using gel electrophoresis.This experiment integrates fundamental knowledge from multiple disciplines,demonstrating the application of this knowledge in research through multi-step experiments.It helps undergraduate students better understand these concepts,combining theory with practice and developing their comprehensive skills.
