摘  要:为了广泛利用海洋微生物发酵产酶制剂,实验室前期筛选出一株具有产高效、高活性外切菊粉酶的海洋放线菌菌株——灰平链霉菌S501。本试验采用单因素试验、筛选因子试验、响应面试验优化其固态发酵产菊粉酶的发酵工艺。结果表明:最佳发酵条件为固态基质选择麸皮,诱导碳源(大蒜皮)添加量10%、诱导氮源(NH_(4)NO_(3))0.05%、NaCl 0.05%、K_(2)HPO_(4)·3H_(2)O 0.05%、MgSO_(4)·7H_(2)O 0.05%、FeSO_(4)·7H_(2)O 0.001%(均以基质麸皮质量计),在接种量8%、培养基固液比(以基质麸皮计,g/mL)1∶1.4、发酵时间141 h、发酵温度28℃、培养基pH值9.0的条件下,外切菊粉酶活力为(209.63±0.96)U/g,该法为工业用微生物法生产酶制剂提供了参考数据。In order to make extensive use of marine microorganisms for fermentation to produce enzyme preparations,the laboratory pre-screened a marine actinomycete strain,streptomyces griseus S501,which has the ability to produce highly efficient and active exo-inulinase.In this experiment,we used the one-way test,the screening factorial test,and the response surface test to optimize the fermentation process of its solid-state fermentation for the production of inulinase.The results showed that the optimal fermentation conditions were as follows:solid substrate selection of bran,10%addition of induced carbon source(garlic peel),0.05%of induced nitrogen source(NH_(4)NO_(3)),0.05%of NaCl,0.05%of K_(2)HPO_(4)·3H_(2)O,0.05%of MgSO 4·7H_(2)O,0.05% of FeSO_(4)·7H_(2)O,and 0.001%of FeSO_(4)·7H_(2)O(all in terms of the mass of substrate bran).Under the optimized conditions of 8%inoculum,medium solid-liquid ratio(in terms of substrate bran,g/mL)1∶1.4,fermentation time 141 h,fermentation temperature 28℃,and medium pH 9.0,the enzyme activity of exo-inulinase was(209.63±0.96)U/g,which provided reference data for the production of enzyme preparations by industrial microbial method.
