摘 要:目的通过皮下注射脱氢表雄酮(DHEA)联合高脂饮食喂养构建多囊卵巢综合征(PCOS)小鼠模型,模拟PCOS的生殖和代谢特点,探索更理想的PCOS小鼠动物模型的构建方法。方法选取30只3周龄的C57BL/6小鼠,随机分为对照组和DHEA联合高脂饮食组(DHEA+高脂饮食组),采用DHEA联合高脂饮食的方式造模21 d,通过对小鼠体质量变化趋势、血清睾酮水平、动情周期、卵巢形态等指标进行模型评价。结果从第4天开始,与对照组比较,DHEA+高脂饮食组小鼠体质量增长明显(P=0.034)。造模完成后,DHEA+高脂饮食组小鼠血清睾酮含量明显升高(P<0.001),动情周期紊乱,长期处于发情期。HE染色结果显示,DHEA+高脂饮食组小鼠的卵巢体积增大,未成熟卵泡数量增多且呈串珠样排列,有较大的囊性卵泡,卵泡内颗粒细胞排列疏松。结论DHEA联合高脂饮食小鼠模型可以更好地模拟PCOS生殖与代谢特征,是一种理想的PCOS动物造模方法。Objective To explore a more ideal method for constructing polycystic ovary syndrome(PCOS)mouse model,PCOS mouse model was constructed by subcutaneous injection of dehydroepiandrosterone(DHEA)combined with high-fat diet feeding to simulate the reproductive and metabolic characteristics of PCOS.Methods Thirty 3-week-old C57BL/6 mice were randomly divided into normal and DHEA combined with high-fat diet group.Through modeling by DHEA injection combined with a high-fat diet for 21 days,model was evaluated by a trend curve of body weight change,serum testosterone,motility cycle,ovarian morphology,and other indexes.Results Starting from the fourth day,the weight of DHEA combined with high-fat diet mice had significantly increased than normal group mice.Upon completion of modeling,serum testosterone were significantly higher in DHEA combined high-fat diet group(P<0.001),and its estrous cycle was disordered in estrus for a long time.HE staining results showed that the ovaries of the mice in the DHEA combined high-fat diet group were enlarged,with an increased number of immature follicles in a bead-like arrangement,larger cystic follicles,and loosely arranged granulosa cells within the follicles.Conclusion DHEA combined with a high-fat diet mouse model could be better simulate reproductive and metabolic characteristics of PCOS,which is an ideal method.
