摘 要:为了研究丁香精油对鸡源超广谱β-内酰胺酶(extended spectrum beta-lactamases,ESBLs)大肠杆菌的抗菌作用,试验采用气相色谱-质谱(gas chromatography-mass spectrometry,GC-MS)方法测定丁香精油成分;采用纸片扩散法测定从信阳市某鸡场病鸡分离的ESBLs大肠杆菌的耐药谱;采用二倍稀释法测定丁香精油对ESBLs大肠杆菌的最小抑菌浓度(MIC)和最小杀菌浓度(MBC);分析丁香精油对ESBLs大肠杆菌β-内酰胺酶活性的影响及耐药性诱导和调节作用。结果表明:丁香精油的主要成分为丁香酚,相对含量为73.88%;试验共鉴定出16株ESBLs阳性大肠杆菌;ESBLs大肠杆菌呈现多重耐药性,其中对5种药物耐药的菌株占6.25%,对6种药物耐药的菌株占31.25%,对7种药物耐药的菌株占31.25%,对8种药物耐药的菌株占25.00%,对9种药物耐药的菌株占6.25%;丁香精油对16株ESBLs大肠杆菌的抑菌圈直径为14.07~23.03 mm;MIC为0.0125%~0.1%,MBC为0.025%~0.2%(V/V)。丁香精油诱导ESBLs大肠杆菌25代后,MIC没有变化;ESBLs大肠杆菌经丁香精油处理后,β-内酰胺酶活性极显著降低(P<0.01),对黏杆菌素、阿莫西林、强力霉素、恩诺沙星、新霉素和庆大霉素的耐药率极显著降低(P<0.01),对氨苄西林的耐药率显著降低(P<0.05)。说明丁香精油对ESBLs大肠杆菌具有良好的抗菌作用,能降低其耐药性,不容易诱导耐药。In order to study the antibacterial effects of clove essential oil against chicken extended-spectrumβ-lactamases(ESBLs)Escherichia coli(E.coli),in the test,gas chromatography-mass spectrometry(GC-MS)was used to determine the composition of clove essential oil.The drug resistance spectrum of ESBLs isolated from sick chickens in a chicken farm in Xinyang City was determined by paper disc diffusion method.The minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of clove essential oil against ESBLs E.coli were determined by two-fold dilution method.The effects of clove essential oil on theβ-lactamase activity of ESBLs E.coli and its induction and regulation effects on drug resistance were analyzed.The results showed that the main component of clove essential oil was eugenol with the relative content of 73.88%.A total of 16 ESBLs-positive E.coli strains were identified.ESBLs E.coli showed multi-drug resistance,including 6.25%of the strains resistant to 5 drugs,31.25%of the strains resistant to 6 drugs,31.25%of the strains resistant to 7 drugs,25.00%of the strains resistant to 8 drugs,and 6.25%of the strains resistant to 9 drugs.The diameters of the inhibition zones of clove essential oil against 16 ESBLs E.coli were 14.07-23.03 mm.The MICs were 0.0125%-0.1%;the MBCs were 0.025%-0.2%.After 25 generations of ESBLs E.coli induced by clove essential oil,there was no change in MIC.The activity ofβ-lactamase was significantly reduced after ESBLs E.coli were treated with clove essential oil(P<0.01);the resistance rates to colistin,amoxicillin,doxycycline,enrofloxacin,neomycin and gentamicin were significantly reduced(P<0.01);the drug resistance rate to ampicillin was significantly reduced(P<0.05).The results indicated that clove essential oil had a good antibacterial effects on ESBLs E.coli,which could reduce its drug resistance and was not easy to induce drug resistance.
