摘  要:试验旨在通过建立红花籽粕中2-羟基牛蒡子苷和罗汉松树脂苷的提取和检测方法,探索鱼产品饲料中红花籽粕的适宜添加量。采用80%乙醇超声提取红花籽粕中2-羟基牛蒡子苷和罗汉松树脂苷,并利用高效液相色谱(HPLC)紫外检测器进行测定。结果表明,2-羟基牛蒡子苷标准液的线性回归方程为:y=10.06885x+0.59591,在2.5~50.0 mg/L范围内线性关系良好,相关系数大于0.9999;在0.5 mg/L添加水平下,平均回收率为94.86%~104.69%,检测限为0.03 mg/L,定量限为0.10 mg/L。罗汉松树脂苷标准液的线性回归方程为:y=15.79781x-17.84546,在2.5~20.0 mg/L范围内线性关系良好,相关系数大于0.9951;在0.5 mg/L添加水平下,平均回收率为99.31%~105.16%,检测限为0.12 mg/L,定量限为0.40 mg/L。研究表明,试验所建立的高效液相色谱法操作简便、准确、重复性好,可应用于红花籽粕中2-羟基牛蒡子苷和罗汉松树脂苷的含量测定。The experiment aims to establish extraction and detection methods for 2-hydroxyarctiin and matairesinoside in safflower seed cake to explore the appropriate addition level of safflower seed cake in fish product feed.The extraction of 2-hydroxy-arctioside and podocarpinus resinoside from safflower seed cake by ultrasonography with 80%ethanol was studied and determined by high performance liquid chromatography(HPLC)ultraviolet detector.The results showed that the linear regression equation for 2-hydroxyarctiin standard solution was y=10.06885x+0.59591.It exhibited a good linear relationship in the range of 2.5~50.0 mg/L,with a correlation coefficient greater than 0.9999.At the addition level of 0.5 mg/L,the average recovery rate was 94.86%~104.69%,with a detection limit of 0.03 mg/L and a quantification limit of 0.10 mg/L.The linear regression equation for matairesinoside standard solution was y=15.79781x-17.84546.It exhibited a good linear relationship in the range of 2.5~20.0 mg/L,with a correlation coefficient greater than 0.9951.At the addition level of 0.5 mg/L,the average recovery rate was 99.31%~105.16%,with a detection limit of 0.12 mg/L and a quantification limit of 0.40 mg/L.The study indicates that the HPLC method established in the experiment is simple,accurate,and has good repeatability.It can be applied to the determination of the content of 2-hydroxyarctiin and matairesinoside in safflower seed cake.
