摘  要:目的探讨外纺锤体极样1(ESPL1)在胰腺腺癌(PAAD)中的表达、分子机制及临床意义。方法整合计算ESPL1信使RNA(mRNA)在1140例PAAD组织和713例对照组织的表达(来源于公共数据库,截至2023年4月31日)。并对47例PAAD和14例对照组织进行免疫组织化学实验。使用Wilcoxon秩和检验和标准化均数差分析ESPL1表达的临床意义。通过基因本体论和京都基因与基因组百科全书富集分析探索PAAD中ESPL1相关的生物学行为。用Q检验和I 2统计分析选择计算ESPL1的SMD的计算模型。结果ESPL1 mRNA在PAAD组织中过表达[SMD=0.19,95%可信区间(CI):0.09~0.28,P<0.05]。内部免疫组织化学验证ESPL1在PAAD中的过表达(Z=-5.62,中位数8.90比2.10,P<0.05)。富集分析表明PAAD中的ESPL1可能与细胞周期等通路有关。结论过表达的ESPL1可能通过细胞周期相关通路参与PAAD的发生发展。Objective To assess the expression status,molecular mechanism,and clinical significance of extra spindle pole bodies like 1(ESPL1)in pancreatic adenocarcinoma.Methods The expression of ESPL1 mRNA in 1140 PAAD tissues and 713 control tissues was integrated and calculated(from public databases,as of April 31,2023).Immunohistochemistry experiments were performed in 47 PAAD and 14 control tissues.The clinical significance of ESPL1 expression was analyzed using Wilcoxon rank sum test and standardized mean difference.Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analysis were used to explore the biological behaviour of ESPL1 in PAAD.The computational model for calculating the SMD of ESPL1 was selected using Q test and I 2 statistical analysis.Results ESPL1 mRNA was overexpressed in PAAD tissues[SMD=0.19,95%confidence interval(CI):0.09-0.28,P<0.05].In-house immunohistochemistry validated the overexpression of ESPL1 in PAAD(Z=-5.62,median 8.90 vs.2.10,P<0.05).Enrichment analyses indicated that ESPL1 in PAAD may be associated with pathways such as cell cycle.Conclusion It was suggested that overexpressed ESPL1 may be involved in the development of PAAD through cell cycle-related pathways.
