摘 要:目的建立电信号调控大鼠运动行为改良实验模型。方法使用脑立体定位仪将3D刺激电极精准植入大鼠颅内的内侧前脑束(MFB)和丘脑腹后中间核(VPM),10~12周龄的无特定病原体(SPF)级雄性Wistar大鼠22只,体重为(320±20)g,购自山西省人民医院实验动物中心;设置未手术组、手术后未刺激组各8只大鼠,手术后刺激组6只,对手术后刺激组6只大鼠实施适宜参数的电信号刺激,完成前进训练和转向测试。再将所有大鼠各进行3次迷宫任务测试,记录手术后刺激组大鼠最佳刺激参数、完成迷宫任务时间等,采用单因素方差分析,组间比较采用LSD法。结果经过训练2~3 d,采用刺激频率60~150 Hz,刺激电压0.6~2.1 V,刺激时长为0.5 s的脉冲对MFB脑区刺激时,安静状态下的大鼠在矿场区域内表现出持续地向前运动。采用刺激频率30~100 Hz,刺激电压0.5~1.8 V,刺激时长为0.5 s的脉冲对VPM脑区进行刺激时,大鼠可出现45°~120°的左右转向。手术后刺激组执行迷宫任务时间[(54.00±11.23)s]低于未手术组[(289.75±136.72)s]、手术后未刺激组[(293.50±149.34)s],3组在时间上差异有统计学意义(F=8.141,P<0.05);多重比较采用LSD检验法,未手术组与手术后未刺激组比较,差异无统计学意义(查t值表,t 19,0.05/2=2.093,LSD-t=-0.061,P>0.05),手术后刺激组执行迷宫任务时间均低于未手术组、手术后未刺激组,差异均有统计学意义(t 19,0.05/2=2.093,LSD-t=3.550,LSD-t=3.600,LSD-t>t 19,0.05/2,P<0.05)。结论采用改良造模方法及适宜电信号刺激参数,可高效率训练出易调控的大鼠运动实验模型。Objective To sstablish an improved experimental model of motor behavior in rats regulated by electrical signals.Methods 3D stimulating electrodes were precisely implanted into the brain of rats using a brain stereotaxic instrument[22 specific pathogen free(SPF)grade male Wistar rats aged 10-12 weeks,weighing(320±20)g,purchased from the Experimental Animal Center of Shanxi Provincial People’s Hospital;8 rats in the non-operation group,8 rats in the non-stimulation group after operation,and 6 rats in the stimulation group after operation]in the medial forebrain bundle(MFB)and ventroposterior intermediate nucleus of thalamus(VPM),and 6 rats in the stimulation group after operation were stimulated with electrical signals of appropriate parameters to complete the forward training and steering test.Then all rats were tested for maze task for three times,and the optimal stimulation parameters and time to complete maze task of rats in the stimulation group after surgery were recorded.The statistical method was one-way ANOVA,and the comparison between groups was performed by LSD method.Results After 2-3 days of training,when MFB brain regions were stimulated with 60-150 Hz stimulation frequency,0.6-2.1 V stimulation voltage and 0.5 s stimulation duration pulses,rats in a quiet state showed continuous forward movement in the mine area.When the VPM brain region was stimulated by a pulse with a stimulation frequency of 30-100 Hz,a stimulation voltage of 0.5-1.8 V,and a stimulation duration of 0.5 s,rats could turn left and right at 45°-120°.The maze task time of the stimulation group after operation[(54.00±11.23)s]was shorter than that of the non-operation group[(289.75±136.72)s]and the non-stimulation group after operation[(293.50±149.34)s].The results of one-way ANOVA showed that the three groups had statistical significance in time(F=8.141,P<0.05);LSD test was used for multiple comparisons.There was no significant difference between the non-operation group and the non-stimulation group after operation(check the t-
