摘 要:目的:探讨白藜芦醇是否能够通过调节TLR4/NF-κB通路来保护视网膜神经节细胞免受急性高眼压导致的死亡。方法:采用小鼠急性高眼压的体内模型。视网膜铺片免疫染色法来评估视网膜神经节细胞的存活,使用蛋白凝胶分析TLR4、NF-κB蛋白的表达,暗适应视网膜电图评定视网膜的功能。结果:与对照组相比,急性高眼压损伤后视网膜神经节细胞呈进行性死亡,而给予白藜芦醇处理后可以明显减少急性高眼压诱导的视网膜神经节细胞死亡。同时结果显示,模型组中视网膜电图的振幅比对照组显著降低,而给予白藜芦醇干预后能够明显阻止视网膜电图振幅的下降。此外,经白藜芦醇治疗后的视网膜损伤组中TLR4、NF-κB蛋白的表达量也比单独损伤组显著下调。结论:白藜芦醇治疗能够有效地保护急性高眼压损伤导致的视网膜神经节细胞死亡和视网膜功能降低,并且可能部分与抑制TLR4/NF-κB通路的活化有关。Objective:To investigate whether resveratrol can modulate TLR4/NF-κB pathway to protect retinal ganglion cells from death elicited by acute ocular hypertension.Methods:An acute ocular hypertension model in mice was utilized in this study.We also used retinal flat-mount immunostaining to assess retinal ganglion cell survival and employed Western blot to evaluate TLR4 and NF-κB protein expression.In addition,scotopic electroretinogram was adopted to estimate retinal function.Results:By comparison with those in the control group,retinal ganglion cells showed progressive death after acute ocular hypertension injury,and treatment with resveratrol significantly reduced retinal ganglion cell death induced by acute ocular hypertension.Meanwhile,the results indicated that the amplitude of retinal electroretinogram in the model groups was markedly lower than that of controls,and resveratrol administration can remarkably prevent the decrease in retinal electroretinogram amplitude.In addition,the protein expression of TLR4 and NF-κB in the retinal injury group treated with resveratrol was also notably downregulated as compared with the injury group alone.Conclusion:Resveratrol administration could effectively reduce retinal ganglion cell death and decrease retinal function elicited by acute ocular hypertension injury,and might be partially related to the inhibition of TLR4/NF-κB pathway activation.
