摘 要:目的:探讨环状RNA hsa_circ_0117530促进宫颈癌发展的机制。方法:通过免疫组织化学方法检测ZEB2在宫颈癌组织中的表达,利用circBase和circInteractome数据库进行分析,筛选出环状RNA hsa_circ_0117530。随后,敲低和过表达hsa_circ_0117530,利用CCK-8法检测宫颈癌细胞增殖活力。此外,使用CircInteractome数据库预测可能与hsa_circ_0117530结合的miRNA并使用StarBase数据库预测miR-874在宫颈癌细胞中的下游靶基因。采用荧光素酶报告基因实验验证并进一步分析其下游的靶点。结果:证实ZEB2在宫颈癌组织中显著上调,发现hsa_circ_0117530分子源自ZEB2基因,并在宫颈癌组织中的表达显著增高。进一步检测结果显示敲低hsa_circ_0117530抑制了宫颈癌细胞系的生长、增殖和迁移,而过表达则产生相反作用。双荧光素酶报告实验表明hsa_circ_0117530与miR-874相互作用,且miR-874直接靶向ZEB1基因。结论:hsa_circ_0117530通过调控miR-874/ZEB1轴促进了宫颈癌细胞的生物功能。Objective:To investigate the mechanisms underlying the promotion of cervical cancer development by circular RNA hsa_circ_0117530.Methods:Immunohistochemistry was used to detect the expression of ZEB2 in cervical cancer tissues.The circBase and circInteractome databases were used to analyze and screen out the circular RNA hsa_circ_0117530.Then,knockdown and overexpression of hsa_circ_0117530 were performed,and the proliferative activity of cervical cancer cells was assessed using the CCK-8 assay.In addition,the CircInteractome database was utilized to predict potential miRNAs that could interact with hsa_circ_0117530,and the StarBase database was utilized to predict the downstream target genes of miR-874 in cervical cancer cells.Luciferase reporter gene experiments were used for verification,and its downstream targets were further analyzed.Results:ZEB2 was significantly upregulated in cervical cancer tissues.The circular RNA hsa_circ_0117530 was identified as originating from the ZEB2 gene,and its expression exhibited a significant upregulation in cervical cancer tissues.Further analysis revealed that knockdown of hsa_circ_0117530 inhibited the growth,proliferation,and migration of cervical cancer cell lines,while overexpression had the opposite effect.Luciferase reporter assays confirmed the interaction between hsa_circ_0117530 and miR-874,with miR-874 directly targeting the ZEB1 gene.Conclusion:The hsa_circ_0117530 promotes the biological functions of cervical cancer cells by regulating the miR-874/ZEB1 axis.
