摘 要:文章旨在优化鸡骨草黄酮提取工艺及鸡骨草黄酮对高脂饲料诱导高脂血症小鼠的降脂减肥作用。采用新型溶剂离子液体-超声波辅助提取鸡骨草黄酮,通过设计响应面Box-Behnken试验优化提取工艺;选取雄性昆明小鼠40只,分为5组:正常对照组、高脂模型组、药物组(阳性药组、鸡骨草提取物低剂量组和高剂量组)。结果表明,鸡骨草黄酮的最佳提取工艺为离子液体浓度1.10 mol/L、乙醇体积分数69.0%、超声时间57 min、料液比1:20(g/mL),得到鸡骨草黄酮得率为(2.950±0.051)%,模型稳定可靠;造模30 d,与正常对照组相比,高脂模型组与药物组小鼠血脂总胆固醇、甘油三酯、低密度脂蛋白含量升高显著(P<0.05),模型建立成功;给予鸡骨草黄酮干预24 d后,与高脂模型组相比,鸡骨草黄酮能够有效控制小鼠体质量增长,显著降低脏器系数和血脂总胆固醇、甘油三酯、低密度脂蛋白、丙二醛含量,升高高密度脂蛋白含量。研究表明鸡骨草黄酮具有调节高脂血症小鼠血脂水平及保护肝脏作用。The aim of this paper was to optimize the extraction technology of flavonoids from Abrus cantoniensis Hance and the effect of Abrus cantoniensis Hance flavonoids on lipid reduction in mice with hyperlipidemia induced by high fat diet.A novel solvent ionic liquid assisted by ultrasonic wave was used to extract the flavonoids from Abrus cantoniensis Hance.Box-Behnken test was designed to optimize the extraction process.Forty male Kunming mice were selected and divided into 5 groups:normal control group,high fat model group,drug group(positive drug group,low dose group,high dose group).The results showed that the optimal extraction process was as follows:ionic liquid concentration of 1.10 mol/L,ethanol concentration of 69.0%,ultrasonic time of 57 min,solid-liquid ratio of 1:20(g/mL),the yield of flavonoids was(2.950±0.051)%,and the model was stable and reliable.After 30 days of modeling,compared with the normal control group,the contents of total cholesterol,triglyceride and low density lipoprotein in high-fat model group and drug group were significantly increased(P<0.05),indicating that the model was successfully established.After 24 days of treatment,compared with the high-fat model group,the Abrus cantoniensis Hance flavonoids can effectively control the growth of body mass,significantly reduce the visceral coefficient,the contents of total cholesterol,triglyceride,low density lipoprotein,malondialdehyde,and increase the content of high density lipoprotein.The study showed that the Abrus cantoniensis Hance flavonoids could regulate the blood lipid level and protect the liver of hyperlipidemia mice.
