摘 要:柞蚕蛹油中不饱和脂肪酸质量分数高达79%,如何高效快速提取柞蚕蛹油、富集其活性成分一直是研究中的重点和难点.本文通过正交试验对超声辅助β-环糊精包合法富集柞蚕蛹油不饱和脂肪酸进行优化,得到最佳试验条件.当超声时间60 min、超声温度65℃、β-环糊精投入量21.5 g时,柞蚕蛹油混合脂肪酸(以α-亚麻酸为主)包合度最高,所得α-亚麻酸质量分数为(39.28±2.57)%.根据傅里叶红外光谱法、高效液相色谱法以及气相质谱法等对富集的不饱和脂肪酸含量及性质进行分析,结果显示,以乙醇作为皂化介质并进行皂化预处理时,柞蚕蛹油特征峰吸光度最大,说明皂化预处理有利于进一步提取不饱和脂肪酸,且未改变蛹油的二级结构,因此超声辅助β-环糊精包合法富集蛹油不饱和脂肪酸可以在试验及生产中应用.The oil from oak silkworm pupae is rich in unsaturated fatty acids(UFAS),which has been a focus of research for and rapid extraction and enrichment of its active components.This study optimizes the ultrasound-assistedβ-cyclodextrin(β-CD)inclusion method foring UFAs from oak silkworm pupae oil using orthogonal experiments.The optimal conditions were found to be 60 minutes of ultrasound time,65℃ultrasound temperature,and 21.5 g ofβ-CD,under which the inclusion rate of mixed fatty acids(primarilyα-linolen acid)from the oak silkworm pupae oil was the highest,with anα-linolenic acid content of(39.28±2.57)%.The content and properties of the enriched UFAs were analyzed using Fourier transform infrared spectroscopy,which showed that the sapon pre-treatment with ethanol as the saponification medium resulted in the highest absorbance peak for the oak silkworm pupae oil,indicating that saponification-treatment facilitates further extraction of UFAs without altering the secondary structure of the pupae oil.Therefore,the ultrasound-assistedβ-CD inclusion method foring UFAs from oak silkworm pupae oil can be applied in both experimental and production settings.
