摘  要:H_(2)S是具有恶臭气味的气体污染物,采用细菌降解H_(2)S具有快捷、安全、成本低等优势。以北海红树林底泥作为菌源,共纯化出4株能降解H_(2)S的菌株,分别为JH2、PS1、PL1与PL2。16S rRNA基因测序结果表明:JH2和PS1菌株均属于芽孢杆菌属,分别与花椒芽孢杆菌(Bacillus zanthoxyli)和巨大芽孢杆菌(Bacillus qingshengii)亲缘关系最为接近;PL1菌株为葡萄球菌属,与沃氏葡萄球菌(Staphylococcus warneri)亲缘关系最为接近;PL2菌株为肠球菌属,与粪肠球菌(Enterococcus faecalis)亲缘关系最为接近。在液体培养基中,4株菌株对H_(2)S的降解率均超过30%,其中,JH2降解率最高,达到54%。进一步对JH2的生长条件进行优化,结果表明:JH2的最适生长条件是pH值为7.0、接种量为15%、Na2S底物质量浓度为0.5 g/L。本研究筛选得到能有效降解H_(2)S的菌株,为生物法治理H_(2)S污染提供了微生物资源。H_(2)S is a gaseous pollutant with a malodorous odor,and the degradation of H_(2)S by bacteria offers the advantages of speed,safety and low cost.In this study,four H_(2)S-degrading bacterial strains,JH2,PS1,PL1 and PL2 were purified from the mangrove substrate of the Beihai Mangrove as the bacterial source.16S rRNA gene sequencing results show that JH2 and PS1 strains belong to the genus Bacillus,which are closest in kinship to Bacillus zanthoxyli and Bacillus qingshengii.PL1 strain belongs to the genus Staphylococcus,which is closest to Staphylococcus warneri.PL2 strain is a genus of Enterococci,which is closest to Enterococcus faecalis.In the liquid medium,all four strains showed more than 30%degradation of H_(2)S,among which,JH2 showed the highest degradation rate of 54%.Further optimization of the growth conditions of JH2 showed that pH 7.0,inoculum amount of 15%and Na2S substrate concentration of 0.5 g/L were the optimal growth conditions for JH2.In this study,the strains that can effectively degrade H_(2)S were screened and provided microbial resources for microbial management of H_(2)S pollution.
