摘 要:目的建立龙眼核标准汤剂高效液相色谱(HPLC)指纹图谱方法,探讨龙眼核标准汤剂抗氧化作用及其与化学成分之间的谱效关系。方法采用HPLC法建立17批不同产地龙眼核标准汤剂指纹图谱,运用相似度分析、聚类分析、主成分分析评价其质量,以铁离子还原能力(FRAP)法测定其总抗氧化能力并通过灰色关联度分析、双变量相关性分析探究发挥抗氧化活性的关键物质。结果17批龙眼核标准汤剂相似度均>0.9,共标定15个共有峰,指认出5个化合物,聚类分析将其聚为两类,主成分分析共提取3个主成分。17批龙眼核标准汤剂均表现出一定的抗氧化能力,总抗氧化能力范围为0.69~1.54 mmol·g^(-1);灰色关联度分析结果表明除F1外,其余共有峰对抗氧化活性均具有一定贡献(关联度值>0.6);双变量相关性分析结果表明,F2、F3、F5~F12、F14与抗氧化活性成显著正相关。结论建立的龙眼核标准汤剂HPLC指纹图谱方法稳定、可靠,17批龙眼核标准汤剂抗氧化活性良好,其发挥抗氧化活性是多组分协同作用的结果。Objective To establish a high performance liquid chromatography(HPLC)fingerprint of longan seed standard decoction,and determine the antioxidant effect of longan seed standard decoction and its spectrum effect relationship in chemical components.Methods The fingerprints of 17 batches of longan seed standard decoction from different origins were established by HPLC.Similarity analysis,cluster analysis,and principal component analysis were used to evaluate its quality,the ferric ion reducing antioxidant power method was used to determine its total antioxidant capacity.The key substances with antioxidant activity were measured by grey correlation analysis and bivariate correlation analysis.Results The similarity of 17 batches of longan seed standard decoction was over 0.9.Totally 15 common peaks were calibrated,5 compounds identified,and clustered into two classes.Three principal components were extracted.The 17 batches of longan seed standard decoction all showed certain antioxidant capacity,with the total antioxidant capacity ranging 0.69~1.54 mmol·g^(-1).The grey correlation analysis showed that except for F1,the other common peaks all contributed to the antioxidant activity(correlation value>0.6),and the bivariate correlation analysis showed that F2,F3,F5~F12 and F14 were positively correlated with the antioxidant activity.Conclusion The HPLC fingerprint of longan seed standard decoction is stable and reliable.All the 17 batches of longan seed standard decoction have good antioxidant activity,its antioxidant with multi-component synergistic effect.
