土茯苓、菊苣、川牛膝对高尿酸血症小鼠降血尿酸作用及机制研究

摘  要:目的研究具有利水祛湿功效的中药土茯苓、菊苣、川牛膝对氧嗪酸钾诱导的高尿酸血症小鼠降尿酸作用及机制。方法48只雄性昆明小鼠按照血尿酸水平分为空白组、模型组、土茯苓组、菊苣组、川牛膝组、别嘌呤醇组,每组8只,采用氧嗪酸钾连续灌胃4周建立高尿酸血症小鼠模型,每日造模后间隔6小时给样,末次给样取血后测定小鼠血尿酸(serum uric acid,SUA)、血尿素氮(blood urea nitrogen,BUN)、血肌酐(serum creatinine,Scr)水平;肝脏中黄嘌呤氧化酶(xanthine oxidase,XOD)、腺苷脱氨酶(adenosine deaminase,ADA);血清中肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-1β(interleukin-1β,IL-1β)含量;回肠组织中三磷酸腺苷结合转运蛋白2(ATP-binding cassette sub-family G member 2,ABCG2)含量。苏木精—伊红(hematoxylin-eosin,HE)染色观察肾组织病理变化。利用蛋白免疫印迹法测定肾脏中ABCG2、有机阴离子转运体1(organic anion transporter 1,OAT1)、尿酸盐阴离子转运体1(recombinant urate transporter 1,URAT1)和葡萄糖转运蛋白9(glucose transporter 9,GLUT9)的蛋白表达。结果通过全自动生化仪检测各组小鼠SUA、BUN、Scr,与模型组比较,各给样组均显著降低;使用酶联免疫吸附法进行检测发现,与模型组比较,各给样组肝脏XOD、ADA、血清TNF-α、IL-1β含量均显著下降(P<0.05),回肠中ABCG2蛋白含量升高;肾组织病理特征明显改善;蛋白免疫印迹结果显示肾脏OAT1和ABCG2蛋白表达显著升高(P<0.05),URAT1和GLUT9蛋白表达有降低趋势。结论具有利水去湿功效的中药土茯苓、菊苣、川牛膝均具有降血尿酸作用,且能够通过抑制炎症因子保护肾脏,推测其作用机制可能与抑制XOD、ADA活性,增加肠道中ABCG2蛋白含量,下调肾脏URAT1、GLUT9蛋白表达,减少重吸收,上调ABCG2、OAT1蛋白表达促进尿酸分泌排泄有关。其中川牛膝抑制血清中炎症因子TNF-α、IL-1β表达效�Objective To study the effect and mechanism of traditional Chinese medicine Glabrous greenbrier rhizome,Chicory and Medical cyathula root,which have the efficacy of eliminating dampness and diuresis,on hyperuricemia mice induced by potassium oxonate.Methods 48 male Kunming mice were divided into control group,model group,Glabrous greenbrier rhizome group,Chicory group,Medical cyathula root group and allopurinol group according to the serum uric acid level,8 mice in each group.The hyperuricemia mouse model was established by continuous gavage of potassium oxonate for 4 weeks,and the samples were given every 6 hours after modeling.After the last sample was given,serum was taken to determine the levels of serum uric acid(SUA),blood urea nitrogen(BUN),serum creatinine(Scr),xanthine oxidase(XOD),liver adenosine deaminase(ADA)in liver,tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)in serum,and ATP-binding cassette sub-family(ATP-2)in ileum.The protein expression of ABCG2,organic anion transporter 1(OAT1),recombinant urate transporter 1(URAT1)and glucose transporter 9(GLUT9)in kidney was determined by protein immunoblotting.Results The levels of SUA,BUN and Scr were detected by fully automated biochemistry in all groups,and compared with the model group,they were all significantly reduced in all administered groups;the detection using enzyme-linked immunosorbent assay found that compared with the model group,the liver XOD,ADA,serum TNF-αand IL-1βcontents in each administered group were all significantly reduced(P<0.05),and the protein content of ABCG2 in the ileum was elevated;the pathological characteristics of renal tissue significantly improved;protein immunoblotting results showed that renal OAT1 and ABCG2 protein expression were significantly increased(P<0.05),URAT1 and GLUT9 protein expression had a tendency to decrease.Conclusions Glabrous greenbrier rhizome,Chicory and Medical cyathula root all have the effect of reducing blood uric acid,and can protect the kidney by inhibiting inflammatory factors
