中药松弛膏外敷联合拉伸训练对废用性肌萎缩大鼠的影响

摘  要:目的 探讨中药松弛膏外敷联合拉伸训练对废用性肌萎缩大鼠腓肠肌组织中相关蛋白表达水平的影响。方法 SPF级Wistar雄性大鼠49只,6周龄,体重190~220 g,随机抽取8只大鼠为正常对照组,采用Nagai法成功建立废用性肌萎缩模型大鼠32只,将其按照随机数字表法分为模型对照组(造模大鼠+自由活动),中药松弛膏干预组(造模大鼠+中药松弛膏外敷),拉伸训练组(造模大鼠+拉伸训练),综合干预组(造模大鼠+中药松弛膏外敷+拉伸训练),每组8只,以上干预均为1次/d,共2周。干预结束后,采用HE染色测量各组大鼠腓肠肌横截面积(CSA)的变化;采用Western blot法和RT-q PCR法检测大鼠腓肠肌组织中肌肉环指蛋白1(MURF1)、肌萎缩盒F因子(MAFbx)、Caspase-3、胰岛素样生长因子1(IGF-1)蛋白及m RNA表达水平。结果 正常对照组肌纤维排列整齐,肌细胞形态正常,无炎症细胞浸润;模型对照组肌纤维排列紊乱,细胞内可见炎症细胞浸润,形态不规则;中药松弛膏干预组肌纤维排列整齐,少量萎缩细胞;拉伸训练组可见少量炎症细胞浸润;综合干预组肌纤维间隙缩小,肌纤维排列整齐,核膜清晰。模型对照组CSA小于正常对照组(P<0.05)。中药松弛膏干预组、拉伸训练组及综合干预组CSA大于模型对照组(P<0.05);综合干预组CSA大于中药松弛膏干预组、拉伸训练组(P<0.05)。模型对照组MURF1、MAFbx、Caspase-3蛋白和m RNA含量较正常对照组升高,IGF-1蛋白和m RNA含量较正常对照组降低(P<0.05)。中药松弛膏干预组和拉伸训练组MAFbx、Caspase-3蛋白和m RNA含量较模型对照组降低(P<0.05);中药松弛膏干预组、拉伸训练组MURF1 m RNA含量较模型对照组降低,IGF-1 m RNA含量较模型对照组升高(P<0.05)。综合干预组MURF1、MAFbx、Caspase-3蛋白和m RNA含量较模型对照组降低,IGF-1蛋白和m RNA含量较模型对照组升高(P<0.05);综合干预组MURF1、MAFbx、Caspase-3蛋白和m RNA含量�Objective To investigate the effect of the external application of Chinese Herbal Relaxation Ointment combined with stretching training on the expression of related proteins in gastrocnemius tissue of disused muscular atrophy rats.Methods There were 49 SPF Wistar male rats,six weeks old and weighing 190-220 g,and eight rats were randomly selected as the normal control group.A total of 32 disused muscular atrophy model rats were successfully established by Nagai method,and were divided into model control group(modeling rats+free movement),Chinese Herbal Relaxation Ointment intervention group(modeling rats+external application of Chinese Herbal Relaxation Ointment),stretching training group(modeling rats+stretching training),and comprehensive intervention group(modeling rats+external application of Chinese Herbal Relaxation Ointment+stretching training)according to random number table method.There were eight rats in each group,and the above intervention was performed once a day for two weeks.After the intervention,the changes of cross sectional area(CSA)of gastrocnemius were measured by HE staining.Western blot and RT-qPCR were used to detect the protein and mRNA expression levels of muscle ring finger 1 (MURF1), muscle atrophy F-box (MAFbx), Caspase-3, and insulin-like growth factor 1 (IGF-1) in rat gastrocnemius tissue. Results In the normal control group, the muscle fibers were arranged neatly, the shape of muscle cells was normal, and there was no inflammatory cell infiltration. In the model control group, the arrangement of muscle fibers was disordered, and inflammatory cells were infiltrated and irregular in shape. In the Chinese Herbal Relaxation Ointment intervention group, the muscle fibers were arranged neatly with a few atrophic cells. A small amount of inflammatory cell infiltration was observed in stretching training group.
