摘 要:目的 探讨半乳糖凝集素-1(Gal-1)表达对增生性瘢痕(HS)组织中TGF-β1/Wnt/β-Catenin信号通路蛋白活性的影响。方法 选择6~8周龄SPF级Gal-1野生型(Gal-1^(+/+))C57BL/6小鼠30只和Gal-1基因敲除(Gal-1^(-/-))C57BL/6小鼠15只(敲除组),雌雄不限,体重60~80 g。按照随机抓取法将Gal-1^(+/+)小鼠分为对照组和模型组,各15只。除对照组外,模型组和敲除组采用恒温恒压电热烫伤法建立HS模型,造模时间为21 d。比较各组瘢痕覆盖率和创口感染率;比较各组成纤维细胞数量和胶原纤维密度;比较各组Gal-1、TGF-β1、Wnt、β-Catenin、Ⅰ型胶原(Col-Ⅰ)和Ⅲ型胶原(Col-Ⅲ)m RNA及蛋白表达量。结果 与模型组比较,敲除组瘢痕覆盖率降低,创口感染率增加(P<0.05)。与对照组比较,模型组成纤维细胞数量和胶原纤维密度增加(P<0.05);与模型组比较,敲除组成纤维细胞数量和胶原纤维密度降低(P<0.05)。与对照组比较,模型组TGF-β1、Wnt、β-Catenin、Col-Ⅰ和Col-Ⅲm RNA及蛋白表达升高(P<0.05);与模型组比较,敲除组TGF-β1、Wnt、β-Catenin、Col-Ⅰ和Col-Ⅲm RNA及蛋白表达降低(P<0.05)。结论 Gal-1高表达可能促进HS中TGF-β1/Wnt/β-Catenin信号通路活性。Objective To investigate the effect of galectin-1(Gal-1)expression on the protein activity of TGF-β1/Wnt/β-Catenin signaling pathway in hypertrophic scar(HS)tissues.Methods Thirty SPF grade Gal-1 wild-type(Gal-1^(+/+))C57BL/6 mice aged 6-8 weeks and fifteen Gal-1 gene knockout(Gal-1^(-/-))C57BL/6 mice(knockout group)were selected,both male and female,weighing 60-80 g.Gal-1^(+/+)mice were divided into control group and model group,with 15 mice in each group.In addition to the control group,the HS model in model group and knockout group was established by constant temperature and constant pressure electrothermal scalding method,and the molding time was 21 d.Scar coverage rate and wound infection rate were compared among all groups;the number of constituent fiber cells and the density of collagen fiber were compared among all groups;and the mRNA and protein expression levels of Gal-1,TGF-β1,Wnt,β-Catenin,typeⅠcollagen(Col-Ⅰ),and typeⅢcollagen(Col-Ⅲ)were compared among all groups.Results Compared with model group,scar coverage rate in knockout group decreased,while wound infection rate increased(P<0.05).Compared with control group,the number of fiber cells and the density of collagen fiber in model group were increased(P<0.05);compared with model group,the number of fiber cells and the density of collagen fiber in knockout group were decreased(P<0.05).Compared with control group,the mRNA and protein expressions of TGF-β1,Wnt,β-Catenin,Col-Ⅰ,and Col-Ⅲin model group were increased(P<0.05);compared with model group,the mRNA and protein expressions of TGF-β1,Wnt,β-Catenin,Col-Ⅰ,and Col-Ⅲin knockout group were decreased(P<0.05).Conclusion High expression of Gal-1 may promote the activity of TGF-β1/Wnt/β-Catenin signaling pathway in HS.
