摘 要:目的 研究阿魏酸(FA)对高糖诱导损伤的人间充质干细胞(MSC)分泌血管生成相关因子及其向血管内皮细胞分化的影响。方法 体外分离、培养并鉴定出人MSC。将得到的人MSC用30 mmol/L的葡萄糖溶液预处理120 h制成高糖诱导损伤人MSC后,分为模型组和实验组,同时设置正常组。用浓度为2 mg/L的FA干预实验组,用等体积的PBS缓冲液干预正常组和模型组。培养48 h后,采用酶联免疫吸附实验法检测各组细胞上清液中VEGF、Ang-1和Ang-2的表达;利用Matrigel成管实验和免疫荧光法分别检测各组人MSC向内皮细胞分化能力及其内皮细胞标志物CD31、v WF的表达水平。结果 成功分离并鉴定出人MSC。与正常组比较,模型组人MSC分泌的VEGF、Ang-1和Ang-2的水平降低,体外成管能力减弱,CD31、v WF的免疫荧光强度降低(P<0.05);与模型组比较,实验组人MSC分泌的血管生成因子VEGF、Ang-1和Ang-2的水平升高,体外成管能力增强,CD31、v WF的免疫荧光强度增强(P<0.05)。结论 阿魏酸具有介导高糖受损人MSC发挥血管新生作用,在糖尿病缺血病变方面具有较高的临床开发应用价值。Objective To investigate the effect of ferulic acid(FA)on the secretion of angiogenesis-related factors and the promotion of differentiation into endothelial cells from human mesenchymal stem cell(MSC)injured by high glacose.Methods MSC were isolated,cultured and identified in vitro.The obtained MSC were pretreated with 30 mmol/L glucose solution for 120 h to make high glucose-induced injury MSC,which were divided into model and experimental groups,while the normal group was set up.FA at a concentration of 2 mg/L was used to intervene in the experimental group,and equal volumes of PBS buffer were used to intervene in the normal and model groups.After 48 hours of culture,the expression levels of VEGF,Ang-1,and Ang-2 in the supernatant from each group were assessed by enzyme linked immunosorbent assay.Additionally,the differentiation capacity towards endothelial cells and the expression of endothelial cell markers CD31 and vWF in MSC were evaluated through Matrigel tube-forming assay and immunofluorescence analysis,respectively.Results Successfully isolated and characterised MSC.Compared with the normal group,the levels of VEGF,Ang-1,and Ang-2 secreted by MSC in the model group were lower,the in vitro tube-forming ability was diminished,and there was a notable reduction in the intensity of immunofluorescence for CD31 and vWF(P<0.05).Compared with the model group,the levels of angiogenic factors VEGF,Ang-1,and Ang-2 secreted by human MSC in the experimental group were increased the ability of tube formation in vitro was enhanced,and the immunofluorescence intensity of CD31 and vWF was enhanced(P<0.05)Results Ferulic acid possesses the capability to modulate the effects of high glucose on MSC,thereby facilitating neovascularization.This compound holds significant promise for clinical application in diabetic ischemic lesions.
