摘　　要：目的 揭示FMNL1对胶质瘤细胞增殖、迁移和侵袭的影响,为胶质瘤患者的临床治疗提供新的潜在作用靶点。方法 利用癌症基因组图谱(TCGA)和中国脑胶质瘤基因组图谱(CGGA)数据库,探讨FMNL1与胶质瘤分级和患者预后之间的联系。通过CCK-8、细胞划痕及Transwell迁移与侵袭细胞功能试验,研究FMNL1对胶质瘤细胞增殖、迁移和侵袭的影响。Western blot(WB)检测U251-shFMNL1、U251-OEFMNL1、U87-OEFMNL1及LN229-OEFMNL1细胞株中上皮间质转化(EMT)的标志物表达水平,初步探讨FMNL1对EMT的影响。结果 临床样本数据库显示FMNL1的表达水平与胶质瘤的WHO级别正相关,并显著影响胶质瘤患者的生存时间。敲低或上调FMNL1的表达对胶质瘤细胞的增殖、迁移和侵袭能力有着显著影响。WB结果显示敲低FMNL1的表达后,E-cadherin的表达显著升高,而N-cadherin、Vimentin的表达显著降低;上调FMNL1的表达后,E-cadherin的表达显著下降,而N-cadherin、Vimentin的表达显著增加。结论 FMNL1的表达水平与胶质瘤WHO级别及胶质瘤患者的预后密切相关。FMNL1作为促癌因子能够显著提高胶质瘤细胞的增殖、迁移和侵袭能力,同时FMNL1促进了胶质瘤细胞的EMT过程。Objective To investigate the effect of FMNL1 on the proliferation,migration and invasion of glioma cells,and to provide a new potential target for the clinical treatment of glioma patients.Methods The Cancer Genome Atlas(TCGA) and Chinese Glioma Genome Atlas(CGGA) databases were used to explore the relationship between FMNL1 and glioma grade and prognosis of patients.The effect of FMNL1 on the proliferation,migration and invasion of glioma cells was investigated by CCK-8,cell scratch and Transwell migration and invasion function experiments.Western blot(WB) was used to detect the epithelial mesenchymal transition(EMT) in U251-shFMNL1,U251-OEFMNL1,U87-OEFMNL1 and LN229-OEFMNL1 cell lines Transition(EMT),and to explore the effect of FMNL1 on EMT.Results The database of clinical samples showed that the expression level of FMNL1 was positively correlated with the WHO grade of glioma and significantly affected the survival time of glioma patients.Knocking down or up-regulating the expression of FMNL1 had a significant effect on the proliferation,migration,and invasive ability of glioma cells.The results of WB showed that knocking down the expression of FMNL1 significantly elevated the expression of E-cadherin expression was significantly elevated,while the expression of N-cadherin and Vimentin was significantly decreased after knocking down the expression of FMNL1.After up-regulating the expression of FMNL1,the expression of E-cadherin was significantly decreased,while the expression of N-cadherin and Vimentin was significantly increased.Conclusions The expression level of FMNL1 is closely related to the WHO grade and the prognosis of glioma patients.As a cancer promoting factor,FMNL1 can significantly improve the proliferation,migration and invasion ability of glioma cells,and at the same time FMNL1 promotes the EMT process of glioma cells.
