摘 要:铁死亡是一种以脂质过氧化及活性氧(ROS)蓄积为特征的新型细胞死亡方式。大量研究表明,铁死亡在多种肿瘤的发生发展过程中发挥重要作用。目前认为,靶向铁死亡可抑制肿瘤生长并增加多种肿瘤对放射治疗及化疗的敏感性,因此诱导铁死亡拓展了肿瘤治疗策略。胶质瘤是成人最常见的脑肿瘤,手术切除联合术后辅助放化疗是其标准治疗策略,然而治疗疗效较差。近年来人们对铁死亡的研究发现其通过抑制谷胱甘肽合成、促进脂质过氧化以调控胶质瘤放疗敏感性。激活铁死亡可能成为药物开发的新靶点,这可能有助于改善胶质瘤患者预后。本文综述了铁死亡在胶质瘤中的最新研究进展及其潜在作用机制,系统讨论了靶向铁死亡提高患者生存期的临床应用前景,旨在为探索胶质瘤治疗的新策略提供理论依据。Ferroptosis is a new type of cell death characterized by lipid peroxidation and reactive oxygen species(ROS) accumulation.A large number of studies show that ferroptosis plays an important role in the occurrence and development of various tumors.It is currently believed that targeting ferroptosis can inhibit tumor growth and increase the sensitivity of various tumors to radiotherapy and chemotherapy.Therefore,inducing ferroptosis expands tumor treatment strategies.Glioma is the most common brain tumor in adults.Surgical resection combined with postoperative adjuvant radiotherapy and chemotherapy is the standard treatment strategy.However,the therapeutic effect is poor.In recent years,research on ferroptosis has found that it regulates glioma radiotherapy sensitivity by inhibiting glutathione synthesis and promoting lipid peroxidation.Activating ferroptosis may become a new target for drugs,which may help improve the prognosis of glioma patients.This article reviews the latest research progress and potential mechanisms of ferroptosis in glioma,and systematically discusses the clinical application prospects of targeting ferroptosis to improve patient survival,aiming to provide a theoretical basis for exploring new strategies for glioma treatment.
