摘  要:硫化氢(H_(2)S)是硫代谢中的重要产物,为探讨硫代谢在二氧化硫(SO_(2))维持鲜食葡萄采后品质中的作用,以及H_(2)S在此过程中的潜在功能,以‘木纳格’葡萄为试材,分别使用500μL/L SO_(2)和0.5 mmol/L H_(2)S抑制剂——DL-炔丙基甘氨酸(DL-propargylglycine,PAG)结合SO_(2)处理。PAG于采摘前24 h喷洒,对照组使用蒸馏水处理。果实在(0±1)℃、相对湿度80%~90%条件下贮藏,每10 d测定硬度、落粒率、腐烂率、硫代谢物质含量、酶活性及相关基因表达水平,并进行相关性分析。结果表明:SO_(2)处理能够维持果实硬度,抑制落粒率和腐烂率的上升。贮藏结束时,SO_(2)处理组果实硬度比对照组高36.9%,落粒率和腐烂率分别比对照组低70.5%和83.7%。同时,SO_(2)促进了硫代谢物质SO_(3)^(2-)、H_(2)S、半胱氨酸和谷胱甘肽(glutathione,GSH)的积累,通过上调硫代谢途径相关编码基因表达提高了亚硫酸盐还原酶、丝氨酸乙酰基转移酶、O-乙酰丝氨酸(硫醇)裂解酶、D-半胱氨酸脱巯基酶和L-半胱氨酸脱巯基酶、胱硫醚β-合酶和3-巯基丙酮酸硫转移酶的活性。上述结果说明SO_(2)对硫代谢具有积极的正调控作用。相关性分析表明,SO_(3)^(2-)、H_(2)S和GSH与果实硬度呈极显著正相关,与落粒率和腐烂率呈显著或极显著负相关。PAG结合SO_(2)处理通过抑制H_(2)S的合成减弱了SO_(2)对于硫代谢的调控,进而降低SO_(2)的保鲜效果。综上所述,SO_(2)能够通过增强硫代谢水平有效维持葡萄采后品质。Hydrogen sulfide(H_(2)S)is a key intermediate in sulfur metabolism.In order to investigate the role of sulfur metabolism in the effect of sulfur dioxide(SO_(2))on maintaining the post-harvest quality of table grapes,as well as the potential function of H_(2)S in this process,‘Munage’grapes(Vitis vinifera L.)were treated with 500μL/L SO_(2)alone or in combination with 0.5 mmol/L DL-propargylglycine(PAG)as a H_(2)S inhibitor.PAG was sprayed on grapes 24 h before harvest and distilled water was used as a control.Grapes were stored at(0±1)℃and 80%-90%relative humidity.Fruit firmness,berry shatter,decay incidence,sulfur metabolites,enzymatic activities and gene expression levels were evaluated every 10 days,and correlation analysis was performed.The results showed that SO_(2)treatment maintained fruit firmness and inhibited the increase of berry shatter and decay incidence.At the end of storage,the fruit hardness of the SO_(2)treatment group was 36.9%higher than that of the control group,and the grain drop rate and decay rate were 70.5%and 83.7%lower than those of the control group,respectively.Additionally,SO_(2)promoted the accumulation of the sulfur metabolites SO_(3)^(2-),H_(2)S,cysteine and glutathione(GSH),and improved the activities of sulfite reductase,serine acetyltransferase,O-acetylserine(thiol)lyase,D-cysteine desulftosylase,L-cysteine desulfhydrylase,cystathionβ-synthase and 3-mercaptopyruvate sulfur transferase by up-regulating the expression of sulfur metabolism pathway-related genes.These indicated that SO_(2)had a positively regulatory effect on sulfur metabolism.Correlation analysis revealed that SO_(3)^(2-),H_(2)S,and GSH were positively correlated with fruit firmness and negatively with shatter and decay rates.The combination of PAG and SO_(2)weakened the regulatory effect of SO_(2)on sulfur metabolism and thereby the preservation effect of SO_(2)by inhibiting the synthesis of H_(2)S.In conclusion,SO_(2)can effectively maintain the postharvest quality of grapes by enhancing sulfur metaboli
