摘 要:为提高阿维菌素发酵水平,文章通过单因素试验筛选发酵培养基中碳源、氮源、前体物质、微量元素和无机盐的最优组分及添加浓度。结果显示:最适主要碳源为糊精,最优添加浓度为120 g/L;最适氮源为黄豆饼粉,最优添加浓度为20g/L;硝酸铵最优添加浓度为0.30g/L;前体物质丙酸钠的最优添加浓度为0.06g/L;微量元素氯化钴、硫酸镍、钼酸钠的最优添加浓度分别为0.020、0.004、0.020 g/L;无机盐磷酸氢二钾和氯化钾的最优添加浓度分别为0.6 g/L和2.0 g/L。在最优培养基配方下,阿维菌素B1a效价较原培养基提高了7.93%。综上所述,文章优化后的发酵培养基可有效提升阿维菌素发酵水平,为阿维菌素的规模化生产奠定基础。In order to improve the fermentation level of avermectin,the optimal composition and concentration of carbon source,nitrogen source,precursor substance,trace element and inorganic salt in fermentation medium were screened by single factor test.The results showed that the optimal main carbon source was dextrin and the optimal additive concentration was 120 g/L,the optimal nitrogen source was soybean cake powder and the optimal additive concentration was 20 g/L,the optimal additive concentration of ammonium nitrate was 0.30 g/L,and the optimal additive concentration of sodium propionate was 0.06 g/L.The optimal addition concentrations of trace elements cobalt chloride,nickel sulfate and sodium molybdate were 0.020,0.004 and 0.020 g/L,respectively.The optimal addition concentrations of inorganic salts dipotassium phosphate and potassium chloride were 0.6 g/L and 2.0 g/L,respectively.Under the optimal medium formulation,the titer of avermectin B1a was increased by 7.93%compared with the original medium.In summary,the optimized fermentation medium in this study can effectively improve the fermentation level of avermectin and lay a foundation for the large-scale production of avermectin.
