摘 要:目的探讨肿瘤蛋白D52(TPD52)基因在乳腺癌组织中的表达水平,分析其与患者临床病理学特征和预后的关系,并进一步探究TPD52在乳腺癌发生发展中可能的生物学功能。方法通过TCGA、GTEx以及在线数据库分析TPD52在乳腺癌组织和正常癌旁组织中的表达水平,同时基于TPD52表达水平的中位数值将乳腺癌患者分为TPD52高表达组及低表达组,研究TPD52表达水平与乳腺癌患者临床参数的相关性。此外,通过Kaplan-Meier方法以及Cox回归探究TPD52与乳腺癌患者预后的相关性。依据基因富集分析探索TPD52的生物学作用。结果与正常组织相比,TPD52在乳腺癌组织中表达升高(P<0.05),与患者的年龄以及M分期密切相关(P<0.05)。生存分析结果显示,TPD52高表达患者总体生存率更低(P<0.05)。功能富集分析显示,TPD52主要参与蛋白质输出、RNA运输、核糖体蛋白复合物结合、糖基磷脂酰肌醇锚生物合成以及同源重组等信号通路。结论TPD52基因在乳腺癌组织中呈高表达状态,且与患者的总体生存率相关,TPD52是乳腺癌发生发展的影响因素之一。Objective To investigate the expression level of TPD52 gene in breast cancer tissues,and to analyze its relationship with clinicopathological features and prognosis of patients,so as to further explore the underlying biological functions of TPD52 in the development of breast cancer.Methods TCGA,GTEx and related online databases were used to analyze the expression levels of TPD52 in breast cancer tissues and normal paracancerous tissues.Breast cancer patients were divided into TPD52 high-expression and low-expression groups according to the median value of TPD52 expression levels.We then investigated the correlation between the expression levels of TPD52 and clinical parameters of breast cancer patients.In addition,the correlation between TPD52 and the prognosis of breast cancer patients was explored by the Kaplan-Meier method and Cox regression.The biological role of TPD52 was explored based on gene enrichment analysis.Results TPD52 expression was significantly elevated in breast cancer tissues compared with normal tissues(P<0.05),and was strongly correlated with the age of the patients as well as the M stage(P<0.05).Survival analysis showed that overall survival was significantly lower in patients with high TPD52 expression(P<0.05).Functional enrichment analysis showed that TPD52 was mainly involved in signaling pathways such as protein export,RNA transport,ribosomal protein complex binding,glycosylphosphatidylinositol(GPI)-anchor biosynthesis,and homologous recombination.Conclusion TPD52 gene is highly expressed in breast cancer tissues and is correlated with the overall survival of patients.Furthermore,TPD52 is one of the factors influencing the development of breast cancer.
