摘 要:菱形蛋白酶属于一种保守的膜内丝氨酸蛋白酶,参与调控植物的生长发育以及对逆境胁迫的响应。对马铃薯RBL基因家族进行全基因组鉴定,利用生物信息学手段对家族成员基因结构、保守基序、顺式作用元件、染色体定位以及基因共线性进行分析,同时结合转录组数据以及qRT-PCR进行表达模式验证。结果表明,马铃薯12个StRBL基因家族成员共分为5个亚族,分布在9条染色体上。位于相同亚族的家族成员的基因结构和保守基序基本一致;顺式作用元件分析发现,马铃薯RBL基因家族成员中含有大量与光反应和逆境胁迫响应有关的顺式作用元件;共线性结果表明,马铃薯与拟南芥、番茄和辣椒存在不同数量的同源基因对,而在烟草中未发现同源基因对;转录组数据和qRT-PCR结果表明,马铃薯RBL基因家族成员在干旱胁迫和盐胁迫条件下高度表达,不同家族成员存在表达差异,StRBL6和StRBL7在干旱胁迫下上调表达,StRBL1、StRBL7、StRBL10和StRBL12在盐胁迫下上调表达,推测RBL基因家族可能参与马铃薯的逆境胁迫响应。研究结果为进一步深入探究马铃薯RBL基因的功能及作用机制奠定了基础。Rhomboid-like(RBL)belongs to a conserved intramembrane serine protease,which plays an important role in plant growth and development and response to adversity stress.In this study,genome-wide identification of the potato RBL gene family was performed and gene structure,conserved motifs,cis-acting elements,chromatogram localization and gene covariance were analyzed by bioinformatics,the expression pattern was verified by combining transcriptome data and qRT-PCR.The results showed that the 12 StRBL gene family members in potato were divided into five subfamilies,distributed on nine chromosomes.The gene structures and conserved motifs of the family members located in the same subfamily were basically the same;the potato RBL gene family members contained a large number of cis-acting elements related to the light response and the response to adverse stress;the results of the covariance showed that there were homologous pairs of genes between potato and Arabidopsis thaliana,tomato,and chili pepper,but not in tobacco;the transcriptome data and qRT-PCR showed that potato RBL gene family members were highly expressed under drought stress and salt stress conditions,and there were expression differences among family members,StRBL6 and StRBL7 were up-regulated under drought stress,StRBL1,StRBL7,StRBL10 and StRBL12 were significantly up-regulated under salt stress.It is speculated that RBL gene family may be involved in potato stress response.The results lay a foundation for further study on the function and mechanism of potato RBL gene.
