摘  要:通过HPLC分析西红花冻干品与烘干品中的三种指标性成分含量差异,为亳州西红花干燥工艺的优化提供支持。采用HPLC法测定西红花中的西红花苷-Ⅰ、西红花苷-Ⅱ、苦番红花素的含量,色谱条件为流动相为甲醇-水,流速为1.0 mL/min,检测波长分别为250 nm(苦番红花素)和440 nm(西红花苷-Ⅰ、西红花苷-Ⅱ),柱温为30℃,进样量为20μL。测定了18批西红花药材(其中2批产自伊朗,其余产自亳州)指标性成分的含量,西红花冻干品中西红花苷-Ι、西红花苷-Ⅱ、苦番红花素的含量平均值分别为51.67μg/mL、11.58μg/mL、27.01μg/mL,与烘干品之间无明显差异。该方法准确、便捷,为西红花质量控制提供了依据,亳州产西红花的指标性成分含量高于伊朗产西红花。冷冻干燥是值得探索的制作高品质西红花的干燥工艺。To analyze the indicative components of freeze-dried and dried saffron with HPLC,provide technical support for the optimization of Bozhou saffron drying process.The contents of crocin I,crocin II and picrocrocin in saffron were determined by HPLC.The chromatographic conditions:mobile phase:methanol-water,flow rate of 1.0 mL/min,detection wavelength of 250 nm(picrocrocin)and 440 nm(crocinⅠ,crocinⅡ),column temperature of 30℃,sample volume of was 20μL.The 3 components of 18 batches of saffron(2 batches from Iran and 16 batches from Bozhou)were determined.The average contents of crocinΙ,crocin II and picrocrocin in freeze-dried saffron were 51.67μg/mL,11.58μg/mL and 27.01μg/mL,respectively,which were close to that in fried saffron.Three components of Bozhou saffron were higher than that of Iran saffron.This HPLC methodwas accurate,convenient,could help the quality control of saffron.Bozhou saffron possessed higher indicative components.The freeze-dried method was potential in producing saffron with high quality and appearance.
