摘  要:目的:本研究拟通过单宁酸(TA)与细胞外基质蛋白间的氢键作用,在牙囊干细胞膜片表面富集TA分子作为膜片保护层,探究TA保护层在体外扩增及植入体内后对膜片中细胞生物活性功能的影响。方法:先制备不同浓度TA处理的牙囊干细胞膜片,运用HE染色查看膜片结构,免疫荧光染色分析相关蛋白表达,细胞增殖实验测细胞活性。通过荧光探针DCFH-DA检测细胞内ROS水平,RT-qPCR探究氧化应激相关因子表达,DPPH法测对外源性ROS清除力。此外,在大鼠体内构建牙周缺损模型,借Micro CT断层扫描、HE及Masson染色评估修复效果。结果:中浓度TA组在维持细胞干性、诱导成骨分化及体内牙周组织再生等方面表现突出,低浓度利于细胞增殖。结论:TA对牙囊干细胞膜片生物活性功能的影响具有浓度依赖性,高浓度的TA处理会抑制细胞活性,适宜浓度的TA保护层可通过调节局部免疫微环境提升细胞膜片的再生潜能。AIM:This study aimed to investigate the effect of TA(Tannic acid)on the properties of dental follicle stem cell(DFSC)membrane sheets and clarify the potential of TA-containing composite cell membrane sheets in repairing periodontal defects.METHODS:DFSC membrane sheets treated with different concentrations of TA were first prepared.Hematoxylin and eosin(HE)staining was used to examine the membrane sheet structure,immunofluorescence staining was employed to analyze the expression of related proteins,and cell proliferation assays were conducted to measure cell viability.The intracellular ROS level was detected by the fluorescent probe DCFH-DA,the expression of oxidative stress-related factors was explored by RT-qPCR,and the scavenging ability of exogenous ROS was measured by the DPPH method.A periodontal defect model was also established in rats,and the repair effect was evaluated by Micro CT,HE and Masson staining.RESULTS:The medium concentration TA group exhibited significant performance in maintaining cell stemness,inducing osteogenic differentiation and in vivo periodontal tissue regeneration,while the low concentration was beneficial for cell proliferation.CONCLUSION:TA-composited DFSC membrane sheets can effectively repair periodontal defects in rats,indicating that the composite membrane sheets possess have potential in periodontal defect repair.
