摘 要:目的探讨蒺藜皂苷D(Terrestrosin D)对人鼻咽癌CNE-2细胞生物学行为及裸鼠CNE-2细胞移植瘤生长的影响。方法采用CCK-8法检测CNE-2细胞活性及筛选后续实验浓度;将CNE-2细胞分为空白对照组和Terrestrosin D处理组(5、10、20μmol/L);EdU染色观察细胞增殖;流式细胞术检测细胞凋亡;细胞成球实验检测干细胞性;蛋白质免疫印迹法检测细胞增殖、凋亡相关蛋白和干细胞标记物的表达;观察Terrestrosin D对人鼻咽癌裸鼠移植瘤生长的影响。结果CCK-8实验结果显示,与空白对照组比较,20、40、80、120μmol/L Terrestrosin D可显著抑制CNE-2细胞活性(P<0.05),后续选用5、10、20μmol/L作为实验浓度。以5、10、20μmol/L Terrestrosin D处理CNE-2细胞,结果显示,与空白对照组比较,10、20μmol/L Terrestrosin D处理组的CNE-2细胞增殖能力、细胞增殖相关蛋白(Ki-67、VEGF)表达、细胞成球率、成球直径、细胞凋亡相关蛋白(Bcl-2)以及干细胞标记物(SOX2、OCT4)的表达均显著降低(P<0.05),而细胞凋亡率、凋亡相关蛋白(Bax、Caspase-3)表达显著升高(P<0.05)。经20μmol/L Terrestrosin D处理可降低鼻咽癌裸鼠移植瘤质量及肿瘤组织中VEGF阳性表达率,提高肿瘤组织的细胞凋亡率,差异均有统计学意义(P<0.05)。结论Terrestrosin D对人鼻咽癌细胞的生长具有显著抑制作用,可促进鼻咽癌细胞凋亡。Objective To explore the effects of Terrestrosin D on biological behaviors of human nasopharyngeal carcinoma CNE-2 cells and growth of transplanted tumors in nude mice.Methods The cell activity was detected and experimental concentration was screened by CCK-8.CNE-2 cells were divided into blank control group and Terrestrosin D treatment groups(5,10,20μmol/L).The cells proliferation was observed by EdU staining.The apoptosis was detected by flow cytometry.The activity of stem cells was detected by cell spheroidization assay.The expressions of cells proliferation and apoptosis-related proteins,and stem cell markers were detected by Western blotting.The effects of Terrestrosin D on the growth of transplanted tumors in nude mice with nasopharyngeal carcinoma were observed.Results The CCK-8 experiment results showed that Terrestrin D at concentrations of 20,40,80,and 120μmol/L significantly inhibited CNE-2 cell activity compared with the blank control group(P<0.05).Subsequently,5,10,and 20μmol/L were selected as experimental concentrations.CNE-2 cells were treated with Terrestrin D at concentrations of 5,10,and 20μmol/L.The results showed that compared with the blank control group,the CNE-2 cell proliferation ability,expression of cell proliferation related proteins(Ki-67,VEGF),cell spheroidization rate,spheroidization diameter,the expression of apoptosis related protein(Bcl-2)and stem cell markers(SOX2,OCT4)were significantly reduced in the 10 and 20μmol/L Terrestrin D treatment groups(P<0.05),while apoptosis rate and expression of apoptosis related proteins(Bax,caspase-3)were significantly increased(P<0.05).The treatment with 20μmol/L Terrestrin D could reduce the quality of transplanted tumors in nasopharyngeal carcinoma nude mice and the positive expression rate of VEGF in tumor tissues,increase the apoptosis rate of tumor tissues.The differences were statistically significant(P<0.05).Conclusion Terrestrosin D can significantly inhibit the growth of human nasopharyngeal carcinoma and promote the apoptosis of
