摘　　要：目的探讨SYT8对结直肠癌患者总生存期(OS)以及结直肠癌细胞迁移和上皮间质转化(EMT)的影响。方法从癌症基因组图谱(TCGA)数据库中提取结直肠癌数据,采用Kaplan-Meier法分析SYT8表达对结直肠癌患者OS的影响,并通过分层分析比较SYT8表达对有无淋巴结转移的结直肠癌患者OS的影响。通过在结直肠癌细胞系KM12和HT29中转染siRNA-SYT8敲低SYT8表达,Transwell法检测转染后结直肠癌细胞迁移能力的变化。Western blotting检测SYT8对EMT相关标记物表达的影响。采用Pearson相关性检验分析SYT8与EMT相关标记物间表达的相关性。结果TCGA数据库分析结果显示,SYT8高表达与结直肠癌患者预后差相关(P<0.05);发生淋巴结转移的患者中,SYT8高表达者的OS更短(P<0.05);SYT8表达降低可以抑制结直肠癌细胞KM12和HT29的迁移能力;SYT8表达降低会引起上皮钙黏蛋白(E-cadherin)表达上调,神经钙黏蛋白(N-cadherin)、波形蛋白(Vimentin)和锌指蛋白(Snail1)表达下调;TCGA数据库提取的结直肠癌数据中,SYT8与编码E-cadherin的基因CHD1表达呈负相关(r=-0.206,P<0.05),而与Snail1表达呈正相关(r=0.118,P<0.05)。结论SYT8高表达与结直肠癌患者的不良预后相关,在有淋巴结转移的结直肠癌患者中,SYT8高表达预示预后差,且SYT8表达降低可以抑制结直肠癌细胞的迁移能力,其机制可能是通过抑制结直肠癌细胞的EMT来实现的。Objective To investigate the effect of SYT8 on the overall survival(OS)of patients with colorectal cancer,and its effect on colorectal cancer cell migration and epithelial mesenchymal transition(EMT).Methods The Kaplan-Meier method was used to analyze the effect of SYT8 expression on OS of colorectal cancer patients in the the Cancer Genome Atlas(TCGA)database,and the effect of SYT8 expression on survival of colorectal cancer patients with or without lymph node metastasis was compared by stratified analysis.Interference with SYT8 expression was achieved by transfection of knockdown lentivirus(siRNA-SYT8)in KM12,HT29 cells.The migration ability of colorectal cancer cells after transfection was detected by Transwell assay.The effect of SYT8 on the expression of markers related to EMT was detected by western blotting.The correlation of expression between SYT8 and EMT-related markers was analyzed by Pearson correlation.Results Analysis of the TCGA online database showed that high SYT8 expression was significantly associated with poor prognosis in colorectal cancer patients(P<0.05).The OS was shorter in patients with high SYT8 expression in patients who developed lymph node metastases(P<0.05).Reduced SYT8 expression inhibited the migratory ability of colorectal cancer cells KM12 and HT29,and inhibited EMT of KM12 cell line,which resulted in the increase of the expression of E-cadherin,and the decrease of the expression of N-cadherin,Vimentin and Snail1.In colorectal cancer data of TCGA database,SYT8 was negatively correlated with the gene CDH1 encoding E-cadherin(r=-0.206,P<0.05)and positively correlated with Snail1 expression(r=0.118,P<0.05).Conclusion High SYT8 expression is associated with poor prognosis in colorectal cancer patients.In colorectal cancer patients with lymph node metastasis,high SYT8 expression predicts poor prognosis.Reducing SYT8 expression inhibits the migration ability of colorectal cancer cells,and the mechanism of which may be by inhibiting the EMT of colorectal cancer cells.
