摘  要:免疫原性细胞死亡(ICD)作为一种特殊的调节性细胞死亡方式,在肿瘤免疫治疗中占据重要地位。本综述着重阐述了ICD的经典途径,包括免疫原性凋亡和坏死。对目前已在临床中应用的抗癌ICD诱导剂以及处于试验阶段的抗癌ICD诱导剂进行了深入的文献分析,从多个维度总结出理想的ICD诱导剂应当具备的特性。同时,深入探讨了肿瘤微环境中ICD激活树突状细胞成熟并引发T淋巴细胞介导的抗肿瘤免疫的分子信号通路,明确了活性氧(ROS)在其中的关键作用。肿瘤内在机制对肿瘤微环境、肿瘤发展及治疗反应影响重大,而ICD为恢复“冷”肿瘤免疫原性带来了希望。深入研究ICD有助于推动肿瘤治疗新策略的开发,具有重要的理论与实践意义。Immunogenic cell death(ICD),as a specific form of regulated cell death,holds a crucial position in tumor immunotherapy.This review emphasizes the classical pathways of ICD,encompassing immunogenic apoptosis and necrosis.The current known clinical application of anticancer ICD inducers and the experimental stage of anticancer ICD inducers were analyzed in depth,and the characteristics of ideal ICD inducers were summarized from multiple dimensions.Simultaneously,the molecular signaling pathways through which ICD activates the maturation of dendritic cells and triggers the T lymphocyte-mediated antitumor immunity in the tumor microenvironment were profoundly explored,clarifying the key role of reactive oxygen species(ROS)therein.The intrinsic mechanisms of tumors exert a significant influence on the tumor microenvironment,tumor development,and treatment responses,and ICD brings hope for restoring the immunogenicity of“cold”tumors.Intensive research on ICD is beneficial for promoting the development of novel strategies for tumor treatment and possesses significant theoretical and practical implications.
