摘　　要：目的 探讨胰腺癌组织Rab3A相互作用蛋白(Rab3IP)表达及其介导的侵袭作用机制。方法 收集2018年5月—2021年6月滁州第一人民医院收治的76例胰腺癌患者组织样本和临床资料,采用免疫组织化学染色检测76例胰腺癌组织和癌旁组织Rab3IP表达,分析Rab3IP表达与胰腺癌临床资料、总体生存率(OS)、无病生存率(DFS)的关系。利用小干扰RNA技术敲低胰腺癌BxPC-3、Capan-1细胞Rab3IP表达,CCK-8检测细胞增殖能力,细胞划痕实验检测细胞迁移活性,Transwell小室检测细胞侵袭活性,Western blot检测细胞内相关蛋白表达。结果 RAB3IP在胰腺癌组织中高表达率为69.7%(53/76),明显高于癌旁组35.5%(27/76),两组比较差异具有统计学意义(χ^(2)=17.839,P<0.001)。Rab3IP表达与胰腺癌患者TNM分期、淋巴结转移、神经侵犯等有关(均P<0.05);Rab3IP表达是胰腺癌总体生存和无病生存的独立预后因素(P均<0.05)。Rab3IP高表达的胰腺癌患者总生存期和无病生存期明显低于RAB3IP低表达患者(P<0.05)。在胰腺癌BxPC-3和Capan-1细胞中,与siNCtrl组比较,siRab3IP组细胞增殖能力、迁移能力和细胞侵袭能力明显减弱,差异具有统计学意义(P<0.05)。siRab3IP组E-cadherin蛋白表达明显高于siNCtrl组,N-cadherin、MMP-2、MMP-9蛋白表达明显低于siNCtrl组,差异有统计学意义(P<0.05)。结论 胰腺癌组织Rab3IP表达上调,Rab3IP可能通过调节上皮间质转化(EMT)参与胰腺癌细胞的迁移和侵袭。Objective To explore the expression of Rab3A interacting protein(Rab3IP)in pancreatic cancer and its invasion mechanism.Methods A total of 76 tissue samples and clinical data of patients with pancreatic cancer admitted to Chuzhou First People`s Hospital were collected.Immunohistochemical staining method was used to detect Rab3IP expression in 76 cases of pancreatic cancer tissues and adjacent tissues,analyzing the relationship between the expression of Rab3IP and clinical data,overall survival(OS),disease-free survival(DFS).Small interfering RNA technology was used to knock down Rab3IP expression in BxPC-3 and Capan-1 cells of pancreatic cancer;CCK-8 was used to detect cell proliferation;cell scratch test was used to detect cell migration activity;Transwell chamber was used to detect cell invasion activity,and Western blot was used to detect the expression of related proteins in cells.Results The high expression rate of RAB3IP in pancreatic cancer tissue was 69.7%(53/76),which was significantly higher than that in the adjacent group 35.5%(27/76),and the difference between the two groups was statistically significant(χ^(2)=17.839,P<0.001).Rab3IP expression was associated with TNM stage,lymph node metastasis and nerve invasion in pancreatic cancer patients(all P<0.05);Rab3IP expression was an independent prognostic factor for overall survival and disease-free survival of pancreatic cancer(all P<0.05).The overall survival and disease-free survival of pancreatic cancer patients with high Rab3IP expression were significantly lower than those with low RAB3IP expression(P<0.05).In BxPC-3 and Capan-1 cells of pancreatic cancer,compared with the siNCtrl group,the proliferation,migration and invasion of cells in the siRab3IP group were significantly reduced(P<0.05).The expression of E-cadherin protein in the siRab3IP group was significantly higher than that in the siNCtrl group,while the expression of N-cadherin,MMP-2,and MMP-9 proteins was significantly lower than that in the siNCtrl group,and the differences were statis
