摘  要:目的:基于中医整体观思想,通过比较被动皮肤过敏模型(PCA)、全身主动过敏模型(ASA)和PCA+ASA新型模型,以期建立更符合中医诊疗特点的荨麻疹动物模型。方法:70只SD大鼠分为血清组、对照组、模型组(PCA组、ASA组、PCA+ASA7d组、PCA+ASA10d组和PCA+ASA15d组)。采用卵白蛋白制备荨麻疹模型,观察大鼠瘙痒次数、蓝斑、IgE、病理及肥大细胞脱颗粒率情况。结果:①瘙痒:模型组瘙痒次数显著多于对照组(P<0.01);②蓝斑:PCA组和PCA+ASA7d组蓝斑率均为100%,蓝斑界线清晰,形状规则;③血清IgE:PCA+ASA 7d组和ASA组显著高于其他组(P<0.01);①病理学:PCA组和PCA+ASA7d组胶原纤维排列紊乱、坏死,伴炎性细胞浸润,组织水肿;③肥大细胞脱颗粒率:模型组均显著高于对照组,其中PCA+ASA7d组最高(P<0.01)。结论:通过结合PCA和ASA,建立了同时模拟整体免疫反应和局部皮肤症状的新型荨麻疹SD大鼠模型。Objective:Based on the holistic view of Chinese medicine,a new animal model of urticaria was established by comparing passive cutaneous anaphylaxis reaction(PCA),active systemic anaphylaxis(ASA),and PCA+ASA experiments.The aim is to establish an animal model of urticaria that is more in line with the characteristics of Chinese medicine treatment.Methods:Seventy SD rats were divided into serum group,blank group,and model group(PCA group,ASA group,PCA+ASA 7 d group,PCA+ASA 10 d group,PCA+ASA 15 d group).The urticaria model was established using ovalbumin.The frequency of itching,blue spots,IgE levels,pathological changes,and the degranulation rate of mast cells in rats were observed.Results:Pruritus:Compared to the blank group,the number of pruritus in the model group was significantly more than in the blank group(P<0.01).②Blue spots:The blue spot rate was 100%in both the PCA group and the PCA+ASA 7 d group,and the blue spots were clearly defined and regular in shape.③Serum IgE:Serum IgE levels were significantly higher in the PCA+ASA 7 d group and ASA group compared to the other groups(P<0.01).@Pathobiology:Disorganized collagen fibrils and necrosis with inflammatory cell infiltration and tissue edema in the PCA and PCA+ASA 7 d groups.Mast cell degranulation rate:The rate of mast cell degranulation was significantly higher in all model groups than in the blank group,with the highest rate in the PCA+ASA 7 d group(P<0.01).Conclusion:By combining PCA and ASA,a novel SD rat model of urticaria was developed that conforms to simulaty both the overall immune response and local skin symptoms.
