摘  要:海藻酸钠(SA)分别与阿拉伯树胶、羧甲基纤维素钠(CMC-Na)、聚乙烯醇(PVA)、明胶和卡拉胶等胶体混合作为复合材料包埋固定化脂肪酶,以确定较好的复合包埋载体,在此基础上再加入戊二醛作为交联剂,研究包埋交联固定化脂肪酶的最佳条件。结果表明:质量分数为0.8%SA和1.2%CMC-Na作为复合载体,在体积分数为0.25%的戊二醛和质量分数为4%的CaCl_(2)溶液中交联固定20 min,获得的固定化脂肪酶活力回收率最高为58.27%,且在重复反应5次后,相对酶活力仍具有76.08%。Sodium alginate(SA)was mixed with colloids such as gum arabic,sodium carboxymethylcellulose(CMC-Na),polyvinyl alcohol(PVA),gelatin and carrageenan respectively as a composite carrier to embed lipase.The cross-linking agent glutaraldehyde was added into the better composite embedding carrier(SA-CMC-Na)to study the optimal conditions of immobilizing lipase by embedding and cross-linking method.The best results was that 0.8% mass fraction of SA and 1.2% mass fraction of CMC-Na were cross-linked and immobilized with the mass fraction of 0.25% glutaraldehyde in 4% mass fraction of CaCl_(2) solution for 20 minutes.The recovery rate of lipase activity was the highest,reaching 58.27%,and after repeating the reaction 5 times,the relative enzyme activity was still 76.08%.
