摘 要:为了得到一种提取发菜多糖较好的工艺,用DEAE-52型离子交换塔对细胞外粗多糖进行了高精度的分离与提纯,并在渗析条件下除去了细分子,从而成功获取了一种酸性多糖(EPS)。进一步通过Sephadex G-100凝胶过滤柱分离和紫外光扫描分析,证实EPS为均一多糖。在多糖的纯化过程中,用DEAE-Cellulose和NaCl浓度梯度相结合的离子交换树脂洗脱。通过对比色谱图中的保留体积、峰高、峰宽以及对称因子等指标,确定了最佳洗脱参数。在本试验装置中,以2mg/mL的NaCl水为洗脱液,流速设定在2.0ml/min,以0.2 mol/L的NaCl为最佳洗脱条件。本文拟以发菜多糖为主要研究对象,采用Box-Behnken试验法,系统研究不同醇沉时间、醇沉时间、乙醇体积倍数等因素对发菜多糖提取率的影响。优选出乙醇含量95%、醇沉24小时、乙醇体积比4倍的醇沉法,得发菜多糖40.24 mg,产率达到6.2%。抗氧化性实验显示,抗氧化活性试验表明发菜多糖对DPPH、羟基自由基有良好的清除能力。In order to obtain a better process for extracting polysaccharides from Nostoc flagelliforme,DEAE-52 ion exchange tower was used to separate and purify extracellular crude polysaccharides with high precision,and the subdivisions were removed under dialysis conditions,so that an acidic polysaccharide(EPS)was successfully obtained.Further separation and ultraviolet scanning analysis by Sephadex G-100 gel filter column confirmed that EPS was a homogeneous polysaccharide.During the purification of polysaccharides,they are eluted with an ion exchange resin combined with DEAE-Cellulose and NaCl concentration gradients.The optimal elution parameters were determined by comparing the retention volume,peak height,peak width,and symmetry factor in the chromatogram.In this experimental setup,2 mg/mL of NaCl water was used as the eluent,the flow rate was set at 2.0 ml/min,and 0.2mol/L NaCl was used as the optimal elution condition.In this paper,the Box-Behnken test method was used to systematically study the effects of different alcohol precipitation time,alcohol precipitation time,ethanol volume multiple and other factors on the extraction rate of Nostoc flagelliforme polysaccharides.The alcohol precipitation method with ethanol content of 95%,alcohol precipitation for 24 hours,and ethanol volume ratio of 4 times was optimized,and 40.24 mg of polysaccharide was obtained,with a yield of 6.2%.The antioxidant activity test showed that the polysaccharide had excellent scavenging ability to DPPH and hydroxyl radicals.
