摘 要:目的 探讨大黄素对甲状腺未分化癌8505c细胞放射敏感性的影响及其作用机制。方法 设立8505c细胞组、8 Gy/min放射组、大黄素组、8 Gy/min放射+大黄素组,上述各组培养48 h。实验结束后,测定各组细胞细胞活力、单克隆形成数目、细胞凋亡、细胞周期、细胞侵袭水平,RT-PCR法及蛋白印迹法测定细胞miR-139-5p、SMC1A mRNA和蛋白水平。结果 与8505c细胞组比较,8 Gy/min放射组、大黄素组、8 Gy/min放射+大黄素组OD值、存活率、单克隆形成数目、穿膜数降低,凋亡率、G2/M期升高(P<0.05),与8 Gy/min放射组、大黄素组比较,8 Gy/min放射+大黄素组OD值、存活率、单克隆形成数目、穿膜数降低,凋亡率、G2/M期升高(P<0.05)。与8505c细胞组比较,8 Gy/min放射组、大黄素组、8 Gy/min放射+大黄素组miR-139-5p表达水平升高(P<0.05);与8 Gy/min放射组、大黄素组比较,8 Gy/min放射+大黄素组miR-139-5p表达水平升高(P<0.05)。与8505c细胞组比较,8 Gy/min放射组、大黄素组、8 Gy/min放射+大黄素组SMC1A mRNA和蛋白水平降低(P<0.05);与8 Gy/min放射组、大黄素组比较,8 Gy/min放射+大黄素组SMC1A mRNA和蛋白水平降低(P<0.05)。结论 大黄素能明显抑制细胞增殖、侵袭,促进细胞凋亡及细胞周期G2/M停滞,增加甲状腺未分化癌8505c细胞放射敏感性;其机制与大黄素可以促进甲状腺未分化癌8505c细胞高表达miR-139-5p进而抑制SMC1A的表达有关。Objective To explore the effect of emodin on radiosensitivity of thyroid undifferen-tiated carcinoma 8505c cells and its mechanism.Methods 8505c cell group,8 Gy/min radi-ation group,emodin group,and 8 Gy/min radiation+emodin group were set up and cultured for 48 hours.After the experiment,cell viability,number of monoclonal formation,cell apop-tosis,cell cycle,and cell invasion level of each group were determined.RT-PCR and Western blot were used to detect miR-139-5p,SMC1A mRNA and protein.Results Compared with 8505c cell group,OD value,survival rate,number of monoclonal formation and number of membrane penetration were decreased in 8 Gy/min radiation group,emodin group,8 Gy/min radiation+emodin group,while apoptosis rate and G2/M stage were increased(P<0.05).Compared with 8 Gy/min radiation group and emodin group,OD value,survival rate,number of monoclonal formation and number of membrane penetration were decreased(P<0.05).In 8 Gy/min+emodin group,OD value,survival rate,number of monoclonal formation and num-ber of membrane penetration were decreased,while apoptosis rate and G2/M stage were in-creased(P<0.05).Compared with 8505c cell group,the expression level of miR-139-5p in 8 Gy/min radiation group,emodin group and 8 Gy/min radiation+emodin group was increased(P<0.05).Compared with 8 Gy/min radiation group and emodin group,Mir-139-5P expres-sion level was increased.The expression level of miR-139-5p in 8 Gy/min radiation+emodin group was increased(P<0.05).Compared with 8505c cell group,SMC1A mRNA and pro-tein expression levels of 8Gy/min radiation group,emodin group and 8Gy/min radiation+emo-din group were decreased(P<0.05).Compared with 8 Gy/min radiation group and emodin group,SMC1A mrna and protein expression levels were decreased(P<0.05).The mRNA and protein expression levels of SMC1A in 8 Gy/min radiation+emodin group were decreased(P<0.05).Conclusion Emodin can significantly inhibit cell proliferation and invasion,pro-mote cell apoptosis and cell cycle G2/M arrest,and increase the radiosensitivity of th
