摘 要:建立一种显色底物法结合高效液相色谱法测定淡豆豉体外凝血酶活性的抑制作用的方法,并探究淡豆豉水煎液对体外凝血酶活性的抑制作用,以期为临床应用与开发提供参考。对显色底物反应中发色底物浓度、孵育时间和孵育温度进行研究,确定最佳反应条件,结合HPLC测定条件,进行方法学考察,并用此方法测定了13种不同产地来源的淡豆豉水煎液对体外凝血酶活性的抑制作用。当反应体系中S-2238凝血酶发色底物浓度为4.0 mg/mL,孵育温度为37℃,孵育时间为20 min时,体系展现出最大凝血酶抑制活性,该方法稳定可靠。通过该方法进一步测定了淡豆豉水煎液的体外凝血酶抑制作用。结果显示淡豆豉水煎液对体外凝血酶活性具有抑制作用,且不同产地来源的淡豆豉水煎液在体外凝血酶抑制率上差异较大,最高抑制率达到了(89.43±2.05)%,其中五批淡豆豉的凝血酶抑制率超过了50%,其产地来源分别是安徽(四批)和四川(一批)。本实验建立的显色底物法结合HPLC法可用于测定淡豆豉水煎液对体外凝血酶活性抑制作用,淡豆豉水煎液对凝血酶活性具有抑制作用,为后续淡豆豉资源的开发利用提供依据与参考。To develop a novel method that utilizes a chromogenic substrate assay along with high performance liquid chromatography to assess the inhibitory effect of Dandouchi on thrombin activity in vitro,the inhibitory effect of Dandouchi on thrombin activitin vitro was investigated to provide a reference for its clinical applications and further development.The study investigated the concentration of chromogenic substrate,incubation time,and temperature during the substrate chromogenic reaction.The optimum reaction conditions were determined.The methodology was investigated in conjunction with the HPLC determination condition.The inhibitory effects of Dandouchi water decoctions from 13 different sources on thrombin activity in vitro were determined using this method.The maximum thrombin inhibitory activity was observed when the concentration of S-2238 thrombin chromogenic substrate was 4.0 mg/mL,the incubation temperature was 37℃,and the incubation time was 20 min.This method was verified to be reliable and stable for measuring the thrombin inhibitory effect of Dandouchi in vitro.The results indicated that Dandouchi has an inhibitory effect on thrombin activity in vitro.The thrombin inhibition rate varied significantly with the source of the Dandouchi,with the highest inhibition rate at(89.43±2.05)%.Five of the tested batches had thrombin inhibition rates greater than 50%.Among them,four batches are from Anhui province and one is from Sichuan province.A method that combines the chromogenic substrate and HPLC techniques was established to evaluate the inhibitory effect of Dandouchi on thrombin activity in vitro.The results showed that Dandouchi can inhibit thrombin activity,which provides a strong foundation for the future development and utilization of Dandouchi resources.
