摘　　要：为探究顺铂(DDP)对溶瘤Ⅱ型单纯疱疹病毒(OH2)的抗病毒效果,验证OH2与DDP序贯给药对肿瘤的疗效,通过qPCR检测OH2在生产细胞Vero中的拷贝数,并收集病毒上清检测病毒滴度。利用CCK8实验验证OH2与DDP共孵育对肿瘤细胞的杀伤效应。同时,建立荷瘤小鼠模型,记录肿瘤大小及小鼠体重,以评估OH2与DDP序贯给药的治疗作用。结果显示,DDP对OH2具有抗病毒作用,但OH2与DDP共孵育仍能降低肿瘤细胞活性,在动物模型中,OH2与DDP序贯给药对肿瘤的治疗效果不劣于单独用药,且延长序贯给药间隔时间可增强抗肿瘤效果。结论表明,尽管DDP对OH2有抗病毒作用,但两者序贯给药仍展现出积极的抗肿瘤疗效。The purpose of this study was to detect the antiviral effect of Cisplatin(DDP)on oncolytic herpes simplex virus type II(oHSV2-hGM-CSF,OH2),and to verify the therapeutic effect of sequential administration of OH2 and DDP on tumor.To this end,the copy number of OH2 in Vero cells was detected by qPCR,and the virus titer was detected by the viral supernatant.CCK8 was used to verify the killing effect of OH2 co-incubated with DDP on tumor cells.A tumor-bearing mouse model was established,tumor size and body weight were recorded,and the effect of sequential administration of OH2 and DDP on tumor therapy was evaluated.Results showed that DDP had antiviral effect on OH2,but the co-incubation of OH2 and DDP still reduced the activity of tumor cells.In animal models,the sequential administration of OH2 and DDP on tumor was no weaker than that of single administration,and prolonged sequential administration interval of the two drugs had better anti-tumor effect.It is concluded that DDP has antiviral effect on OH2,but sequential administration of the two drugs still has positive antitumor effect.
