摘 要:目的通过构建与糖酵解相关的circRNA介导的ceRNA网络,探索ESCC中的关键基因及其潜在的治疗靶点,为预后标志物的筛选和免疫治疗提供新线索。方法本研究综合分析了GEO、TCGA和GTEx数据库的数据,鉴定了差异表达的circRNA、miRNA和mRNA,并构建了circRNA-miRNA-mRNA的ceRNA网络。使用生存分析确定与ESCC预后密切相关的基因,并通过免疫细胞浸润分析探索这些基因与免疫治疗潜在关系。结果ceRNA网络中的糖酵解相关基因HOMER1的高表达与ESCC患者的高生存率相关,且HOMER1表达与CD4^(+)T细胞的负相关性提示了其在免疫治疗中的潜力。结论本研究揭示了糖酵解相关ceRNA网络在ESCC发病机制中的重要作用,为预后标志物的鉴定和免疫治疗策略的发展提供了新的见解和潜在的研究方向。Objective To identify the key genes and potential therapeutic targets in esophageal squamous cell carcinoma(ESCC)by constructing a glycolysis-related circRNA-mediated ceRNA network,thereby providing new insights for screening prognostic biomarkers and the development of immunotherapy.Methods This study integrated data from the GEO,TCGA,and GTEx databases to identify differentially expressed circRNAs,miRNAs and mRNAs.A circRNA-miRNA-mRNA ceRNA network was constructed.Survival analysis was used to determine genes closely related to ESCC prognosis and immune cell infiltration analysis was conducted to explore the potential relationship between these genes and immunotherapy.Results The high expression of the glycolysis-related gene HOMER1 in the ceRNA network was associated with higher survival rates in ESCC patients.Additionally,the negative correlation between HOMER1 expression and CD4^(+)T cells suggested its potential role in immunotherapy.This study also identified the hsa_circ_0051799/hsa-miR-4710/HOMER1 regulatory axis,highlighting its significance in ESCC.Conclusion This study reveals the significant role of the glycolysis-related ceRNA network in the pathogenesis of ESCC,providing new insights and potential research directions for the identification of prognostic biomarkers and the development of immunotherapeutic strategies.
