转运蛋白在细菌吸附、解毒重金属中的功能预测与分析

摘  要:为研究转运蛋白在细菌吸附、解毒重金属中的作用,以一株从重金属污染土壤中分离出的细菌为研究对象,分别检测了菌株细胞组分对40~160 mg/L的Cu^(2+)、Zn^(2+)、Cd^(2+)和Cr(Ⅵ)的吸附能力,并分析了菌株基因组中与重金属吸附、解毒相关的转运蛋白。结果表明,菌株对120 mg/L的Cd^(2+)的吸附能力最强,吸附率为93.4%。对4种重金属的吸附方式主要是表面吸附,其次对Cu^(2+)和Zn^(2+)为胞外沉淀,对Cd^(2+)和Cr(Ⅵ)为胞内积累。在菌株基因组中,ABC转运蛋白家族的蛋白质参与重金属的转运和吸附,包括磷酸盐转运家族PstSPstBCA、MlaF和MlaE等。NRAMP家族中的MntH蛋白、ZIP家族中的ZupT蛋白因结构中存在大量金属离子结合位点,可为菌株对重金属的转运和胞内积累提供载体。外排系统中的2个CDF家族蛋白、10个RND蛋白和5个P-ATPase蛋白因其特殊结构域通过对金属离子的外排实现菌株对重金属的解毒。To study the role of transporters in adsorption and detoxification of heavy metals by bacteria,the bacteria isolated from heavy metal contaminated soil was studied.The adsorption properties of 40-160 mg/L of Cu^(2+),Zn^(2+),Cd^(2+)and Cr(Ⅵ)were investigated,and the transporters related to the adsorption and detoxification of heavy metals were analyzed.The results showed that the adsorption capacity of 120 mg/L of Cd^(2+)was the strongest(adsorption rate was 93.4%).Furthermore,the four heavy metals were mainly adsorbed by surface adsorption,followed by extracellular precipitation for Cu^(2+)and Zn^(2+),while intracellular accumulation for Cd^(2+)and Cr(Ⅵ).In the strain genome,proteins belonging to ABC transporter family participated the transport and biosorption of heavy metals,including the phosphate transport family PstSPstBCA,MlaF and MlaE.The MntH protein in the NRAMP family and ZupT protein in the ZIP family could provide a carrier for the transport and intracellular accumulation of heavy metals due to the presence of a large number of metal ion binding sites in their structures.In addition,two CDF family proteins,ten RND proteins and five P-ATPase proteins were involved in the efflux of metal ions and detoxification by bacteria due to their special domains.
