摘  要:为探究动物双歧杆菌乳亚种XLTG11对洛哌丁胺构造小鼠便秘模型的缓解作用及机制。将100只健康Balb/c雄性小鼠随机平分为5组,分别为空白组、模型组、动物双歧杆菌乳亚种XLTG11低、中、高剂量组。通过观察粪便基础指标、组织学形态、短链脂肪酸和微生物群组成分等比较不同剂量动物双歧杆菌乳亚种XLTG11对便秘小鼠的影响以及缓解机制。结果表明,动物双歧杆菌乳亚种XLTG11各剂量组均能显著增加小鼠首粒排黑便时间、5 h粪便粒数、粪便含水率和小肠推进率;下调促炎因子,上调抗炎因子;升高兴奋型神经递质,降低抑制型神经递质;修复受损结肠屏障;改善肠道菌群,增加有益菌丰度。表明动物双歧杆菌乳亚种XLTG11具有润肠通便作用,其润肠通便机制是通过改善肠道菌群、提高便秘小鼠粪便中乙酸含量进而影响胃肠调节肽和炎性细胞因子释放。文章为拓展益生菌研究方向、开发新型缓解便秘功能性乳制品提供新思路。To investigate the alleviating effect and mechanism of Bifidobacterium animalis subsp.XLTG11(XLTG11)in a loperamide-constructed mouse constipation model.In the article,100 healthy Balb/c male mice were randomly and equally divided into five groups:blank,model,and XLTG11 low,medium,and high dose groups.The effects of different doses of XLTG11 on constipated mice and the mechanism of alleviation were compared by observing fecal basal indexes,histological morphology,short-chain fatty acids,and microbiota composition scores.The results showed that all dose groups of XLTG11 significantly increased the time to the first black stool,the number of fecal pellets in 5 h,the water content of feces,and the rate of small intestinal propulsion in mice;down-regulated pro-inflammatory factors and up-regulated anti-inflammatory factors;elevated excitatory neurotransmitters and lowered inhibitory neurotransmitters;repaired the damaged colonic barrier;and improved the intestinal flora and increased the abundance of beneficial bacteria;XLTG11 was shown to have a laxative effect,and its laxative mechanism may be related to the improvement of the intestinal flora and the increase in the content of acetic acid in the feces of constipated mice,which in turn affects the release of gastrointestinal regulatory peptides and inflammatory cytokines.This study provides new ideas for expanding the research direction of probiotics and developing new functional dairy products for constipation relief.
