基于网络药理学和分子对接探讨鸡血藤治疗绒毛膜下血肿的作用机制

摘  要:目的基于网络药理学与分子对接技术探讨鸡血藤治疗绒毛膜下血肿的作用机制。方法使用TCMSP数据库并筛选确定鸡血藤活性成分,采用SwissTargetPrediction预测鸡血藤的靶点。在GeneCards、OMIM、TTD和DRUGBANK数据库中检索疾病靶点。将两者交集靶点导入Metascape数据库,进行基因本体(GO)和京都基因与基因组百科全书(KEGG)富集分析。通过Cytoscape软件将结果可视化,筛选关键成分和疾病靶点进行分子对接预测。结果共筛选出24个活性成分、131个交集靶点。通过Cytoscape软件筛选出AKT1、IL6、TNF、EGFR、SRC等关键靶点。根据KEGG富集分析,鸡血藤主要通过PI3K-AKT信号通路、EGFR酪氨酸激酶抑制剂抵抗、内分泌抵抗、FAK信号通路等信号通路抑制绒毛膜下血肿的发生发展。根据分子对接验证,AKT1、IL6、TNF、EGFR、SRC与相应的上述关键药物活性成分具有良好的结合能力。结论鸡血藤可能通过活性成分作用于AKT1、IL6、TNF、EGFR、SRC等关键靶点,产生促进血管生成修复、调节机体免疫、调控滋养细胞的侵袭和迁移、改善血栓状态等作用。Objective To explore the mechanism of action of Spatholobus suberectus in treating subchorionic hematoma(SCH)based on network pharmacology and molecular docking techniques.Methods The active components of Spatholobus suberectus were identified using the TCMSP database,and targets were predicted using SwissTargetPrediction.Disease targets were retrieved from GeneCards,OMIM,TTD,and DRUGBANK databases.The intersecting targets were imported into the Metascape database for Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis.Cytoscape software was used to visualize the results,and key components and disease targets were selected for molecular docking prediction.Results A total of 24 active components and 131 intersecting targets were identified.Key targets such as AKT1,IL6,TNF,EGFR,and SRC were selected using Cytoscape software.According to KEGG enrichment analysis,Spatholobus suberectus mainly inhibits the occurrence and development of SCH through pathways such as the PI3K-Akt signaling pathway,EGFR tyrosine kinase inhibitor resistance,endocrine resistance,and the FAK signaling pathway.Molecular docking validation showed that AKT1,IL6,TNF,EGFR,and SRC have good binding abilities with the corresponding key active components of the herb.Conclusion Spatholobus suberectus may exert its therapeutic effects on SCH by acting on key targets such as AKT1,IL6,TNF,EGFR,and SRC,promoting angiogenesis and repair,regulating immune responses,controlling trophoblast invasion and migration,and improving thrombotic status.
