摘 要:目的研究肝癌组织中赖氨酸脱甲基酶2A(KDM2A)、前梯度蛋白2(AGR2)的表达与肝癌患者临床病理特征和生存预后的关系。方法选取2019年1月至2021年11月在本院接受根治治疗的147例肝癌患者,采用免疫组化法检测患者肝癌组织及相应癌旁组织KDM2A、AGR2表达;采用免疫吸附法检测血清甲胎蛋白(AFP)水平;经过门诊复诊或电话对患者进行为其3年的术后随访;采用Spearman法分析肝癌组织中KDM2A、ACR2表达之间的关系;使用多因素Cox回归模型分析影响肝癌患者预后的影响因素;采用Kaplan-Meier分析KDM2A、ACR2与肝癌患者预后的关系。结果昊KDM2A与AGR2在肝癌组织中的阳性表达占比高于二者在癌旁组织中的阳性表达占比(P<0.05),二者在肝癌组织中的表达呈现正相关趋势(P<0.05);KDM2A、ACR2阳性表达、阴性表达的患者在肿瘤直径、肿瘤分期、肿瘤分化程度、淋巴结是否转移以及AFP水平方面具有统计学差异(P<0.05);随访期间共有58例患者死亡,死亡率为39.46%,其中KDM2A阴性表达生存率(44/53,83.02%)高于其阳性表达生存率(45/94,47.87%),ACR2阴性表达生存率(36/41,87.81%)高于其阳性表达生存率(53/106,50.00%)(P<0.001);肿瘤直径≥5cm、肿瘤分期Ⅲ期、淋巴结转移、KDM2A与AGR2阳性表达均是影响肝癌患者生存预后的因素(P<0.05)。结论KDM2A、AGR2在肝癌患者癌组织中阳性表达占比更高,其表达情况与肝癌患者肿瘤直径、肿瘤分期、淋巴结是否转移以及AFP水平有关,其中KDM2A阴性表达生存率为83.02%,ACGR2阴性表达生存率为87.81%,均高于二者阳性表达生存率。Objective To investigate the relationship between the expression of lysine-specific demethylase 2A(KDM2A)and anterior gradient-2(AGR2)in liver cancer tissues and the clinical pathological features and survival prognosis of liver cancer pa-tients.Methods From January 2019 to November 2021,147 liver cancer patients who received radical treatment in our hospital were gathered.Immunohistochemistry was used to detect the expression of KDM2A and AGR2 in liver cancer tissues and coresponding adja-cent tissues of patients.Immunoadsorption method was used to detect serum alpha fetoprotein(AFP).After outpatient follow-up or phone call,the patients were followed up for 3 years after surgery.Spearman method was used to analyze the relationship between KDM2A and AGR2 in liver cancer tissues.Multi factor Cox regression model was used to analyze the influencing factors on prognosis of liver cancer patients.Kaplan-Meier method was used to analyze the relationship between KDM2A,AGR2 and the prognosis of liver cancer patients.Results The proportions of positive expression of KDM2A and ACR2 in liver cancer tissues were higher than those in adjacent tissues(P<0.05),and the expression of the two showed a positive correlation trend in liver cancer tissues(P<0.05).There were statistically obvious differences in tumor diameter,tumor stage,tumor differentiation degree,lymph node metastasis,and AFP between patients with positive and negative expression of KDM2A and AGR2(P<0.05).During follow-up,a total of 58 patients died,with a mortality rate of 39.46%.Among them,the survival rate of KDM2A negative expression(44/53,83.02%)was higher than that of the positive expression(45/94,47.87%),and the survival rate of AGR2 negative expression(36/41,87.81%)was higher than that of the positive expression(53/106,50.00%)(P<0.001).Tumor diameter≥5 cm,tumor stage III,lymph node metastasis,and positive expression of KDM2A and ACR2 were all factors that affected the survival prognosis of liver cancer patients(P<0.05).Conclusion KDM2A and AGR2 have a high
