摘　　要：目的分析PDZ结合激酶(PBK)在肝细胞癌(HCC)组织中的表达及其预后意义,为临床诊治提供潜在标志物。方法收集2020年1月—2023年6月钦州市第二人民医院50对HCC及癌旁正常组织手术切除标本的石蜡切片及对应的临床资料,采用免疫组化法检测PBK表达水平,并分析其与患者临床病理特征及生存预后的关系。结果PBK在HCC组织中的阳性表达率为56.0%,高于癌旁正常组织的34.0%(P<0.05);不同肿瘤大小、转移、临床分期的HCC患者PBK表达情况比较,差异有统计学意义(P<0.05);PBK高表达的患者中位生存期为9.214个月,短于PBK低表达患者的11.030个月,是HCC的独立预后因素(P<0.05);Kaplan-Meier生存曲线显示,PBK高表达患者的1年生存率为32.1%,低于低表达患者的63.6%(P<0.05);COX单因素分析结果显示,PBK表达水平与患者的总体生存率相关(P<0.05)。结论PBK在HCC组织中呈高表达,与不良预后相关,可能成为HCC治疗的新分子靶点及预后评估指标。Objective To analyze the expression of PDZ binding kinase(PBK)in hepatocellular carcinoma(HCC)and its prognostic significance,so as to provide potential markers for clinical diagnosis and treatment.Methods Paraffin sections and corresponding clinical data of 50 pairs of HCC and paracancer normal tissue specimens treated in the Second People's Hospital of Qinzhou from January 2020 to June 2023 were collected,the expression level of PBK was detected by immunohistochemical method,and the relationship between PBK and clinicpathological features and survival prognosis of patients was analyzed.Results The positive expression rate of PBK protein in HCC tissues was 56.0%,which was significantly higher than 34.0%in adjacent normal tissues(P<0.05);the expression of PBK in HCC patients with different tumor size,metastasis and clinical stage was statistically significant(P<0.05);the median survival time of patients with high PBK expression was 9.214 months,which was shorter than 11.030 months of patients with low PBK expression,and was an independent prognostic factor for HCC(P<0.05);Kaplan-Meier survival curve showed that the 1-year survival rate of 32.1%in patients with high PBK expression was lower than 63.6%in patients with low PBK expression(P<0.05);the results of COX univariate analysis showed that PBK expression was correlated with the overall survival of patients(P<0.05).Conclusions PBK is highly expressed in HCC tissues,which is associated with poor prognosis,and may be a new molecular target and prognostic evaluation indicator for HCC.
