血清circ-ATAD1和miR-140-3p表达与宫颈癌患者预后相关性的研究

摘　　要：目的分析血清环状RNA-ATAD1(circ-ATAD1)和微小RNA-140-3p(miR-140-3p)表达与宫颈癌患者预后的相关性。方法选择2018年3月至2020年3月河北省沧州中西医结合医院146例宫颈癌患者(研究组)、146例子宫良性病变患者(子宫良性病变组)及146例体检健康者(对照组)为研究对象。采用实时荧光定量PCR检测血清miR-140-3p、circ-ATAD1水平。采用Kaplan-Meier法分析血清circ-ATAD1、miR-140-3p表达与宫颈癌患者预后的相关性。多因素Cox回归模型分析宫颈癌患者预后的影响因素。结果研究组血清circ-ATAD1水平高于对照组、子宫良性病变组(P<0.05),miR-140-3p水平低于对照组、子宫良性病变组(P<0.05)。有阴道浸润、有淋巴结转移、FIGO分期Ⅲ~Ⅳ期的宫颈癌患者miR-140-3p低表达、circ-ATAD1高表达者占比分别高于无阴道浸润、无淋巴结转移、FIGO分期Ⅰ~Ⅱ期的患者(P<0.05)。血清circ-ATAD1高表达的宫颈癌患者3年生存率低于circ-ATAD1低表达患者(30.14%vs.64.38%,P<0.001)。血清miR-140-3p高表达的宫颈癌患者3年生存率高于miR-140-3p低表达患者(61.64%vs.32.88%,P<0.001)。FIGO分期、circ-ATAD1、淋巴结转移、miR-140-3p为宫颈癌患者预后的影响因素(P<0.05)。结论宫颈癌患者血清circ-ATAD1水平升高而miR-140-3p水平降低,二者与宫颈癌患者淋巴结转移、FIGO分期密切相关,为宫颈癌患者预后的影响因素。Objective To analyze the correlation between serum circular RNA-ATAD1(circ-ATAD1)and microRNA-140-3p(miR-140-3p)expression and the prognosis of cervical cancer patients.Methods From March 2018 to March 2020,a total of 146 patients with cervical cancer(study group),146 patients with benign uterine lesions(benign uterine lesions group),and 146 healthy people who underwent the physical examination(control group)in Cangzhou Hospital of Integrated TCM-WM·Hebei were selected as the research subjects.Real-time quantitative PCR was used to detect serum levels of miR-140-3p and circ-ATAD1.The Kaplan-Meier method was used to analyze the correlation between serum circ-ATAD1 and miR-140-3p expression and the prognosis of cervical cancer patients.Multivariate Cox regression model was used to analyze the influencing factors of prognosis of cervical cancer patients.Results The serum circ-ATAD1 level in the study group was significantly higher than those in the control group and benign uterine lesion group(P<0.05),and miR-140-3p was significantly lower than those in the control group and benign uterine lesion group(P<0.05).The proportions of patients with low expression of miR-140-3p and high expression of circ-ATD1 in the cervical cancer patients with vaginal infiltration,lymph node metastasis,and FIGO stageⅢ-Ⅳwere higher than those in the cervical cancer patients with no vaginal infiltration,no lymph node metastasis,and FIGO stageⅠ-Ⅱ(P<0.05).The 3-year survival rate of cervical cancer patients with high circ-ATD1 expression was lower than that of patients with low circ-ATD1 expression(30.14%vs.64.38%,P<0.001).The 3-year survival rate of cervical cancer patients with high miR-140-3p expression was higher than that of patients with low miR-140-3p expression(61.64%vs.32.88%,P<0.001).FIGO stage,circ-ATAD1,lymph node metastasis and miR-140-3p were factors affecting the prognosis of cervical cancer patients(P<0.05).Conclusion The serum level of circ-ATAD1 in patients with cervical cancer is significantly increased and the l
