大黄素对肝硬化大鼠肝功能及肠黏膜屏障损伤的改善作用

摘  要:目的探讨大黄素通过调节炎症因子表达对肝硬化大鼠肝功能和肠黏膜屏障损伤的改善作用。方法将36只大鼠随机分为正常组、模型组和大黄素组,每组12只,除正常组外,其余组大鼠利用四氯化碳(CCl4)-玉米油复制大鼠肝硬化模型,2次/周,一共12周。造模成功后分别给予大黄素(大黄素组)和生理盐水(模型组)干预,每日1次,连续4周。通过HE、Sirus Red染色分别观察大鼠肝、肠组织的病理结构变化;检测血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、总胆红素(TBIL)、α-羟基丙酸(α-LA)、二胺氧化酶(DAO)和血浆内毒素(ET)水平和回肠组织中白细胞介素(IL)-6、肿瘤坏死因子-α(TNF-α)、IL-17水平;蛋白印迹法检测肠组织中Toll样受体4(TLR4)、核因子-κB(NF-κB)蛋白表达水平。结果与正常组比较,模型组大鼠出现肝细胞坏死、排列紊乱、肠黏膜绒毛脱落及炎症因子浸润等病理变化;与模型组比较,大黄素组肝组织和肠组织中炎症及损伤程度均得到改善,血清中AST、ALT、TBIL、IL-6、TNF-α、IL-17、α-LA、DAO和ET水平及TLR4、NF-κB蛋白表达水平均降低(P<0.05)。结论大黄素能够通过抑制炎症因子释放,减轻炎症反应,从而改善肝功能,降低α-LA、DAO和ET水平,缓解肠黏膜屏障损伤。Objective To investigate the improvement effect of rhodopsin on the hepatic function and intestinal mucosal barrier damage in cirrhotic rats by regulating the expression of inflammatory factors.Methods Thirty-six male rats were randomly divided into normal group,model group and rhodopsin group,12 rats in each group.Except for the normal group,the rats in the remaining groups were used to induce rat cirrhosis model by carbon tetrachloride(CCl4)-corn oil twice/week for a total of 12 weeks.After successful modeling,rhodopsin(rhodopsin group)and saline(model group)interventions were given once daily for 4 weeks,respectively.The pathological and structural changes in rat liver and intestinal tissues were observed by HE and Sirus Red staining.Serum levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBIL),alpha hydroxypropionic acid(α-LA),diamine oxidase(DAO),and plasma endotoxin(ET)and interleukin(IL)-6,tumor necrosis factor alpha(TNF-α),and IL-17 in ileal tissues were detected.Western blot was used to detect the expression levels of Toll like receptor 4(TLR4)and nuclear factor kappa B(NF-κB)proteins in intestinal tissue.Results Compared with normal group,the model group showed pathological changes such as hepatocyte necrosis,disordered arrangement,shedding of intestinal mucosal villi,infiltration of inflammatory factors and so on.Compared with the model group,inflammatory factors and the degree of injury in liver and intestinal tissue of rats in the rhodopsin group were improved,and the serum levels of AST,ALT,TBIL,IL-6,TNF-α,IL-17,α-LA,DAO,ET levels and TLR4 and NF-κP protein expression levels were significantly reduced(P<0.05).Conclusion Rhodopsin is able to attenuate the inflammatory response by inhibiting the release of inflammatory factors,attenuate the inflammatory response thereby improving liver function,decreasingα-LA,DAO,and ET levels,and alleviating intestinal mucosal barrier damage.
