摘  要:目的 通过生物信息学探索分析KCNQ1OT1与肿瘤免疫特征的相关性。方法 通过TISCH数据库中的单细胞测序数据分析KCNQ1OT1和肿瘤免疫微环境的联系。通过Sangerbox 3.0、ACLBI和TIDE数据库中的TCGA、GEO数据分析KCNQ1OT1在泛癌中与肿瘤免疫的相关性。结果 单细胞测序分析显示KCNQ1OT1的表达在多种免疫细胞中异常增加,TCGA数据显示KCNQ1OT1与免疫细胞浸润、免疫评分和免疫检查点的表达均呈负相关关系。此外,TIDE数据库数据显示当KCNQ1OT1在黑色素瘤中高表达时,免疫治疗效果较差,并且可能途径是抑制了细胞毒性T细胞的活性。结论 KCNQ1OT1与抑制性免疫微环境密切相关,可能是预测免疫疗法疗效的生物标志物和新的分子靶点。Objective To explore and analyze the correlation between KCNQ1OT1 and tumor immune characteristics via bioinformatics.Methods Analyze correlation between KCNQ1OT1 and tumor immune microenviron-ment were using single-cell sequencing data from the TISCH database.And the correlation between KCNQ1OT and tumor immunity in pan-cancer were analyzed using TCGA and GEO data from Sangerbox 3.0,ACLBI,and TIDE databases.Re-sults Single-cell sequencing analysis showed that the expression of KCNQ1OT1 was abnormally increased in various im-mune cells.TCGA data indicated that KCNQ1OT1 expression was negatively correlated with immune cell infiltration,im-mune scores,and the expression of immune checkpoints.In addition,TIDE database data showed that when KCNQ1OT1 is highly expressed in melanoma,immunotherapy is less effective.And it may be due to the inhibition of cytotoxic T cell activity.Conclusion KCNQ1OT1 is closely related with an inhibitory immune microenvironment.KCNQ1OT1 may be a biomarker and new molecular target for predicting the immunotherapy efficacy.
