摘  要:为探索鲍曼不动杆菌基因组及分子生物学特征,以临床分离得到的1株病原菌(BM5)为目标菌株,采用药敏试验分析其耐药情况,对其基因组进行高通量测序分析,通过相关数据库及生物信息学方法分析预测其多位点序列分型(MLST)、耐药基因和毒力基因等,并通过分析比较基因组序列相似性,确定其与其他鲍曼不动杆菌菌株在进化上的相关性。结果表明:该菌全基因组为3990385 bp长度的环状DNA,GC含量为38.9%。该菌株还含有2个环状质粒(长度分别为72258和8731 bp)。该菌MLST分型为ST2,共携带85个耐药基因,其中包含耐碳青霉烯类耐药基因blaOXA-167(96.63%),同时,还携带83个毒力基因和5个前噬菌体;该菌可通过多种途径来获得和维持其多耐药性和复杂的致病性。综上,对耐碳青霉烯类鲍曼不动杆菌BM5的基因组特征进行了初步分析,为研究耐碳青霉烯类鲍曼不动杆菌的耐药形成机制及致病机制奠定基础。In order to explore the genome and molecular biological characteristics of Acinetobacter baumannii,a clinically isolated pathogen was selected as the target strain and named BM5.The drug resistance of Acinetobacter baumannii was analyzed by drug sensitivity test,its genome was analyzed by high-throughput sequencing,its multi-site sequence typing(MLST),drug resistance genes and virulence genes were analyzed and predicted by relevant databases and bioinformatics methods,and its evolutionary correlation with other Acinetobacter baumannii strains was determined by analyzing and comparing genomic sequence similarity.The results showed that the whole genome was 3990385 bp circular DNA with GC content of 38.9%.The strain also contained two ring plasmids(72258 and 8731 bp in length).The MLST type of the bacterium was ST2,carring 85 drug resistance genes and including carbapenem resistance genes(96.63%),83 virulence genes and 5 prophages.Therefore,the bacterium may acquire and maintain its multi-drug resistance and complex pathogenicity through multiple pathways.In conclusion,the genomic characteristics of the new carbapenem-resistant Acinetobacter baumannii BM5 were analyzed,which will lay a foundation for further research on the mechanism of drug resistance and pathogenic mechanism of carbapenem-resistant Acinetobacter baumannii.
