摘 要:本文以油茶优良无性系‘赣州油10号’为研究对象,通过探究油茶种子适宜的生活力检测方法和贮藏条件,以期为油茶种子科学研究、种苗生产及反季节育苗提供现实指导。本试验采用3因素4水平正交设计进行油茶种子生活力测定条件优化试验,开展播种前不同处理措施对油茶种子发芽的影响研究,运用TTC染色法对不同贮藏条件下的油茶种子生活力进行比较分析。结果表明:染色时间是影响TTC染色试验结果的最主要因素,油茶种子生活力的最佳检测条件为“1.0%TTC染色浓度+35℃染色温度+24h染色时间”,其油茶种子生活力检测结果为80.0%;播种前药剂种类、处理浓度措施是影响油茶种子发芽的主要因素,在“GA3+0.1%处理浓度+2h处理时间”条件下发芽率最优(77.1%);采用不同方法贮藏油茶种子其生活力差异显著,在4℃、-20℃条件下贮藏180d后种子生活力保持效果较好,常温和真空包装贮藏效果最差。In this article,the excellent clone of Camellia oleifera'Ganzhouyou 10’was taken as the research object and the suitable viability detection methods and storage conditions of C.oleifera seeds was explored to provide practical guidance for scientific research,seedling production and off-season breeding of C.oleifera seeds.In this experiment,a three-factor and four-level orthogonal experiment was used to optimize the conditions for determining the viability of C.oleifera seeds.The research on the impact of different pretreatment measures on the germination of C.oleifera seeds before sowing was carried out.The TTC staining method was used to compare and analyze the viability of C.oleifera seeds under different storage conditions.The results showed that the staining time was the most important factor affecting the results of the TTC staining test.The best detection condition for the viability of C.oleifera seeds was“1.0%TTC staining concentration+35^staining temperature+24 h staining time”,and the detection result was 80.0%.The types of agents and treatment concentration measures before sowing were the main factors afecting the germination of C.oleifera seeds.The germination rate was the best(77.1%)under the treatment condition of“GA3+0.1%treatment concentration+2 h treatment time”.There were significant differences in the viability of C.oleifera seeds stored by different methods.Under the storage temperature treatment conditions of 4 C and—20 C,the seed viability was maintained well after 180 days.The storage effects of normal temperature and vacuum packaging were the worst.
