摘 要:目的 研究不同剂量的放射治疗对新型小鼠坐骨神经癌痛模型的神经病理性疼痛行为的影响。方法 选取体重为18~20 g的BALB/c雌性小鼠24只,采用随机数字表法分为4组:对照组(Ctrl组)、6 Gy X射线治疗组(6 Gy组)、10 GyX射线治疗组(10 Gy组)、15 Gy X射线治疗组(15 Gy组)。测量4组小鼠基础体重、左侧大腿体积及左侧足底机械缩足阈值(MWT)。将结肠癌细胞CT26接种于小鼠坐骨神经旁,每天记录体重、肿瘤体积及MWT。接种后第9天,Ctrl组不进行X线照射,其余组分别接受对应剂量X线照射,每天记录肿瘤体积、MWT以及瘫痪日期。结果 与Ctrl组比较,6 Gy组肿瘤体积增长速度显著减缓(P<0.01),MWT的低峰日期和异常增高日期和出现患肢瘫痪日期显著延迟(P<0.01);与6 Gy组比较,10 Gy组肿瘤体积增长速度进一步减缓(P<0.01),MWT的低峰日期以及异常增高日期显著延迟,出现患肢瘫痪日期延迟(P<0.05);与10 Gy组比较,15 Gy组肿瘤体积增长速度最慢(P<0.01),MWT治疗后未见显著降低,未观察到明显低峰,且未出现MWT异常增高,仅在治疗后第23天1只出现患肢瘫痪,出现患肢瘫痪日期差异无统计学意义(P>0.05)。结论 本研究成功建立小鼠坐骨神经癌痛模型,10 Gy X射线是较理想的放射治疗射线剂量。Objective To investigate the effects of different intensities of radiotherapy on neuropathic pain in a new sciatic nerve cancer pain model in mice.Methods Twenty-four BALB/c female mice weighing 18-20 g were selected,and all were randomly divided into four groups using the random number table method:a con-trol group(Group Ctrl),a 6-gray radiotherapy group(Group 6 Gy),a 10-gray radiotherapy group(Group 10 Gy),and a 15-gray radiotherapy group(Group 15 Gy).The basal body weights,volumes of the left thighs,and bases of the left paws'mechanical withdrawal threshold(MWT)were recorded for each group.Colon cancer cells CT26 were inoculated near the sciatic nerve of each mouse,and data were recorded on consecutive days.On day 9,no radiotherapy was given to Group Ctrl.Radiotherapy of Group 6 Gy intensity was given to Group 6 Gy,10 Gy intensity to Group 10 Gy,and Group 15 Gy intensity to Group 15 Gy,and the dates were recorded on consecutive days.Results Compared with the Group Ctrl,the tumour volume growth rate in the Group 6 Gy was significantly decreased(P<0.01),the date of low peak and abnormal increase in MWT significantly delayed,and the date of appearance of paralysis of the affected limb significantly delayed(P<0.01);compared with Group 6 Gy,the tumour volume growth rate in Group 10 Gy was significantly decreased(P<0.01),the date of low peak and abnormal increase in MWT significantly delayed,the date of the appear-ance of affected limb paralysis delayed(P<0.05);compared with Group 10 Gy,the growth rate of tumour volume in Group 15 Gy was significantly decreased(P<0.01),MWT was not significantly reduced after treat-ment,no obvious low peak was observed,and no abnormal increase in MWT observed,and only one mouse was found to have developed limb paralysis on the 23rd day after treatment,and the date of the appearance of limb paralysis was not significantly different(P>0.05).Conclusion Results from pain behavioural studies sug-gest that the new sciatic nerve cancer pain model in mice has been successfully established an
